陕西枣疯病发病规律与苗木带病检测 被引量:6

摘要 从彬县、佳县和清涧县采集的枣树苗、酸枣苗、枣树昆虫样品中提取总核酸,克隆与测序。结果表明,陕西彬县、佳县和清涧县3个枣区的枣疯病(JWB)植原体16S rDNA基因大小为1.2 kb,基因序列基本一致,为同一个种,JWB和酸枣(WJWB)植原体16S rDNA的亲缘关系达到99.9%。由于JWB与16S rⅤ组B亚组成员榆树黄化(Elm yellows,EY)和悬钩子丛枝病(Rubus witches’broom,RuS)的同源性分别是98.5%和98.3%。因此,JWB归属于植原体16S rⅤ组B亚组。PCR检测苗木结果显示,市场上的枣树苗带病较多,以佳县枣树苗的带病率最高,达到7.69%,彬县带病率最低,为4.35%;清涧县酸枣苗的带病率最高,达到19.05%。对枣疯病品种抗病性调查结果显示,‘婆枣’发病率较高(25.32%),‘晋枣’发病率最低(11.18%)。对在彬县与佳县枣园采集的中华拟菱纹叶蝉(Hishimonoides chinensis)、凹缘菱纹叶蝉(Hishimonus sellatus)、大青叶蝉(Cicadella viridis)和条沙叶蝉(Psammotettix striatus),用PCR检测带毒情况,结果显示,只有中华拟菱纹叶蝉和凹缘菱纹叶蝉携带枣疯病植原体,说明这两种叶蝉是陕西的枣疯病的媒介昆虫。在温室内,采用菟丝子和嫁接方式,能够传播枣疯病。 Jujube witche’s broom(JWB),caused by a phytoplasma,is a devastating disease of jujube in China,causing great economic loss.The total nucleic acids were extracted from the samples of jujube stocks,wild jujube seedlings,insect pests from jujube-productive counties of Binxian,Jiaxian and Qingjianxian in Shaanxi,and Cloning and sequencing of 16S rRNA gene was conducted to detect pathogenic phytoplasma.The results show that the sequences of all 16S rRNA gene are the same 1.2 kb,the identity of 16S rRNA gene is 99.9%between wild jujube witches’broom(WJWB)and the JWB.Based on the identity of JWB 16S rRNA sequence with elm yellows(98.3%)and rubus witches’broom(98.5%),JWB are attributable to 16Sr V group B subgroup.PCR detection showed that jujube stocks carrying JWB were prevalent,with the highest rate of 7.69%in Jiaxian,and the lowest rate of 4.35%in Binxian.Meanwhile,the highest rate of 19.05%was detected in wild jujube in Qingjianxian.The higher incidence of JWB was detected in Pozao jujube(25.32%),and the lowest incidence in Jinzao(11.18%).The PCR screening four leafhoppers,including Hishimonoides chinensis,Hishimonus sellatus,Cicadella viridis,and Psammotettix striatus,only Hishimonoides chinensis and Hishimonus sellatus collected from Binxian and Jiaxian was checked out JWB phytoplasma.The greenhouse trials indicated that JWB phytoplasma could be transmitted by dodder from jujube to periwinkle,also graft in periwinkle.
