喷墨全包芯骨支架材料的制备及生物安全性研究 被引量:3

Preparation and biosafety of a ink-jet all-core structure composite scaffold for bone tissue engineering

摘要 目的观察喷墨全包芯骨支架材料亲水性、组织相容性和生物安全性。方法支架通过快速成型方法制作。亲水性通过吸水率评价;细胞在支架上的增殖能力通过噻唑蓝(MTT)比色法测定,将材料植入大鼠体内,2、4、8、12周进行血常规、肝功能、肾功能检测,观察局部伤口情况及大鼠肝肾功能等指标改变。结果两组材料2、6、10、14 d后MTT比色法测定值分别为0.14、0.28、0.62、0.78和0.13、0.19、0.36、0.46,喷墨全包芯骨支架亲水性明显高于聚乳酸-乙醇酸(PLGA)/磷酸三钙(TCP)骨支架,血常规、肝功能、肾功能检测差异无统计学意义(P>0.05)。结论喷墨全包芯骨支架材料具有良好的亲水性、生物相容性和生物安全性。 Objective To observe the hydrophilicity,biocompatibility and biosafety of ink-jet all-core structure composite scaffold.Methods The ink-jet all-core structure composite scaffold composed of poly(lactic-co-glycolic acid)/β-tricalciumphosphate(PLGA/β-TCP)skeleton ink-jet-wrapped with type I collagen was manufactured via rapid prototyping,and PLGA/β-TCP scaffold was chosen as the control group.Hydrophilicity was evaluated by water absorption rate.The proliferation ability of cells on the scaffold was determined by methyl thiazol tetrazolium(MTT)assay(thiazole blue).The scaffolds were implanted into rats,and blood routine,liver function and renal function tests were performed at 2,4,8,12 weeks,to observe the changes of local wounds and liver and kidney functions in rats.Results For MTT assay,the values at 2nd,6th,10th,and 14th day in two groups were 0.14,0.28,0.62,0.78,and 0.13,0.19,0.36,and 0.46,respectively.The hydrophilicity of the ink-jet all-core structure composite scaffold was significantly higher than that of the PLGA/TCP scaffold.there were no significant differences in blood routine,liver function and renal function between two groups(P>0.05).Conclusion The ink-jet all-core structure composite scaffold possesses admirable hydrophilicity,biocompatibility and biosafety.
