皮层肌动蛋白络氨酸磷酸化对宫颈鳞状上皮细胞癌发展研究

摘要 目的探讨皮层肌动蛋白表达及其磷酸化在宫颈鳞状上皮细胞癌发生、发展过程中的作用。方法选取11例正常宫颈组织、61例宫颈不典型增生组织及116例宫颈鳞状上皮细胞癌组织标本,探讨皮层肌动蛋白和磷酸化皮层肌动蛋白的临床病理学意义,通过体外实验观察皮层肌动蛋白对宫颈鳞状上皮细胞癌细胞的增殖、迁移和侵袭能力的改变。结果与正常宫颈组织比较,磷酸化皮层肌动蛋白的表达在不典型增生及鳞状上皮细胞癌组织显著增高,差异有统计学意义(P<0.05)。宫颈鳞状上皮细胞癌细胞通过干扰皮层肌动蛋白的表达,使细胞增殖、迁移、侵袭能力显著降低,差异有统计学意义(P<0.05)。结论皮层肌动蛋白及磷酸化皮层肌动蛋白的下调,可显著降低宫颈鳞状上皮细胞癌细胞的增殖、迁移和侵袭能力。 Objective To investigate the role of cortactin(CTTN)and phosphor-CTTN(p-CTTN)in the cervical squamous cell carcinoma(SCC)progression.Methods The implication of CTTN and p-CTTN expression were investigated in 11 normal cervix,61 cervical intraepithelial neoplasia(CIN),and 116 cervical SCC tissues by immunohistochemistry.The influence of CTTN expression on proliferation ability,motility and invasion ability of cervical SCC cell lines were investigated in vitro.Results The p-CTTN expression was significantly increased in CIN and cervical SCC than normal cervical tissues(P<0.05).Furthermore,the proliferative,migratory and invasive abilities of the cells were significantly reduced in cervical SCC cells(P<0.05).Conclusion The downregulation of cortactin and phosphorylated cortactin can significantly reduce the proliferation,migration and invasion ability of cervical squamous cell carcinoma cells.
