摘要 目的:探讨降低miR-21表达对顺铂(DDP)耐药A549细胞药物敏感性的影响。方法:采用qRT-PCR对顺铂敏感和耐药的NSCLC患者癌组织、A549及顺铂耐药A549(A549/DDP)细胞中miR-21的表达水平进行检测。利用脂质体转染法将miR-21抑制剂(anti-miR-21)、阴性对照(anti-miR-NC)转染到A549/DDP中,检测顺铂对细胞的IC50,MTT法检测细胞增殖活性,Annexin V-FITC/PI双染法检测细胞凋亡,分光光度法检测Caspase-3活性。结果:与顺铂敏感肿瘤组织和A549细胞相比,顺铂耐药肿瘤组织和A549/DDP中miR-21表达水平升高(P <0. 05)。与anti-miR-NC转染的A549/DDP细胞比较,顺铂对anti-miR-21转染的A549/DDP细胞IC50和细胞增殖活性降低(P <0. 05),细胞凋亡率和Caspase-3活性升高(P <0. 05)。结论:抑制miR-21可提高肺癌细胞对顺铂的敏感性。 Aim: To investigate the effects of inhibiting miR-21 on the sensitivity of cisplatin(DDP)-resistant A549 cells to DDP and its mechanism. Methods: qRT-PCR was used to detect the expression of miR-21 in sensitive or insensitive lung cancer tissue and A549 cells. The inhibitor of miR-21(anti-miR-21),miRNA inhibitor negative control(anti-miRNC) were transfected into DDP-resistant lung cancer cells(A549/DDP) under the action of Lipofectamine 2000. IC50 of DDP to the cells was detected. Cell proliferation activity and apoptosis were measured by MTT and Annexin V-FITC/PI staining,respectively,and Caspase-3 activity was determined by spectrophotometric method. Results: The expression of miR-21 in DDP-resistant tumor tissue or A549/DDP was significantly higher than that in DDP-sensitive tumor tissue or 549 cells(P <0. 05). The IC50 of DDP to anti-miR-21 transfection A549/DDP cells and proliferation activity were lower,while the apoptosis rate and Caspase-3 activity in anti-miR-21 transfection group were higher than those in anti-miR-NC transfection group(P < 0. 05). Conclusion: Inhibition of miR-21 enhances the sensitivity of lung cancer cells to DDP.
