自噬相关蛋白EPG5调节的鼻咽癌细胞经电离辐射后自噬机制的研究 被引量:1

摘要 目的研究电离辐射后鼻咽癌细胞CNE1及CN E2中自噬相关蛋白EPG5的表达情况以及EPG5的表达与鼻咽癌细胞凋亡之间的关系。方法鼻咽癌CNE1及CNE2细胞株接受不同剂量电离辐射后,用WesternBlot法检测自噬相关蛋白EPG5的表达。si RNA转染法特异性敲除epg 5基因后,Annexin V-FITC/PI法检测两株细胞凋亡情况。结果经不同剂量电离辐射后,CNE1细胞株中EPG5的表达量与放射剂量呈正相关,CNE2细胞株中呈负相关。敲除epg 5基因后,EPG 5表达减少的同时,CNE1细胞株凋亡率明显增加,而CNE2细胞株的凋亡率明显降低。结论鼻咽癌细胞经过电离辐射后,EPG5的表达可能与肿瘤细胞的分化程度有关,分化程度较低的细胞表达下调,分化程度较高的细胞表达上调。同时分化程度低的细胞EPG5低表达可以减少细胞凋亡,这可能是低分化鼻咽癌产生放疗抵抗的分子机制。 OBJECTIVE To study the expression of autophagy associated protein EPG5 in nasopharyngeal carcinoma cells CNE1 and CNE2 after ionizing radiation and the relationship between EPG5 expression and apoptosis in nasopharyngeal carcinoma cells. METHODS Different doses of ionizing radiation on nasopharyngeal carcinoma CNE1 and CNE2 cells were carried out,and the autophagy associated protein EPG5 expression was detected by western blot. After specific knockout of EPG5 gene by siRNA transfection method,apoptosis of the two kinds of cells was detected with Annexin V-FITC/PI method. RESULTS After management with different doses of ionizing radiation,the expression of EPG5 in CNE1 cell line was positively correlated with radiation dose,but negatively correlated with radiation dose in CNE2 cell line. When EPG5 was knocked out,the expression of EPG5 was decreased,while the apoptosis rate of CNE1 cell line was increased and that of CNE2 cell line was decreased. CONCLUSION The expression of EPG5 in nasopharyngeal carcinoma cells after ionizing radiation may be related to the differentiation degree of tumor cells. The expression of EPG5 in cells with lower differentiation degree is down-regulated and that in cells with higher differentiation degree is upregulated. At the same time,low expression of EPG5 in poorly differentiated cells can reduce apoptosis,which may be the molecular mechanism of radioresistance in poorly differentiated nasopharyngeal carcinoma.
