雌二醇对高眼压大鼠眼压、视网膜厚度及凋亡、自噬基因表达的调节作用

摘要 目的观察雌二醇(E_2)对高眼压大鼠眼压、视网膜厚度及凋亡、自噬基因表达的调节作用。方法2016年3月—2018年6月在广东省医学实验动物中心进行动物实验,选择雄性SD大鼠24只以随机数字表法分为对照组、高眼压组、E_2组各8只。造模前及造模后1~4周,测定眼压水平;造模后4周,测定视网膜厚度、视神经纤维厚度以及视网膜内凋亡基因、自噬基因的表达水平。结果造模后1~4周,高眼压组和E_2组眼压水平均明显高于对照组,而E_2组眼压水平与高眼压组比较无明显差异(F/P=72. 229/0. 000、69. 970/0. 000、60. 454/0. 000、54.175/0.000)。造模后4周,3组的视网膜厚度、视神经纤维厚度以及视网膜中Bcl-2的mRNA表达量比较,均为高眼压组<E_2组<对照组(F/P=11.062/0. 000、10. 353/0. 000、12. 716/0. 000),视网膜中Bax、Caspase-9、Caspase-3、mTORC1、LC3-Ⅱ、Beclin-1的mRNA表达量比较,均为高眼压组> E_2组>对照组(F/P=86. 916/0. 000、38. 083/0. 000、41.306/0.000、 42. 364/0.000、31.703/0.000、41.232/0.000)。结论雌二醇对高眼压大鼠的眼压水平无明影响,但是能够增加视网膜厚度、抑制视网膜内的细胞凋亡及自噬。 Objective To observe the effects of estradiol(E2) on intraocular pressure, retinal thickness, apoptosis and autophagy gene expression in rats with high intraocular pressure. Methods From March 2016 to June 2018, animal experiments were conducted in Guangdong Medical Laboratory Animal Center. Twenty-four male SD rats were randomly divided into control group, high intraocular pressure group and E2 group with 8 rats in each group. The intraocular pressure(IOP)was measured before and 1-4 weeks after modeling, and the retinal thickness, the thickness of optic nerve fibers, the expression of apoptotic genes and autophagic genes in retina were measured 4 weeks after modeling. Results After 1 to 4 weeks after modeling, the intraocular pressure of the high intraocular pressure group and the E2 group was significantly higher than that of the control group, while the intraocular pressure level of the E2 group was not significantly different from that of the high intraocular pressure group(F/P = 72.229/0.000, F/P = 69.970/0.000, F/P = 60.454/0.000, F/P = 54. 175/0.000). At 4 weeks after model establishment, the retinal thickness, optic nerve fiber thickness, and Bcl-2 mRNA expression in the retina were all in the high intraocular pressure group < E2 group < control group(F/P = 11.062/0.000, F/P = 10.353/0.000,F/P = 12.716/0.000), the mRNA expression levels of Bax, Caspase-9, Caspase-3, mTORC1, LC3-II and Beclin-1 in the retina were all high intraocular pressure group > E2 group > control group(F/P = 86. 916/0. 000, F/P = 38. 083/0. 000, F/P=41.306/0.000,F/P=42.364/0.000,F/P=31.703/0.000,F/P=41.232/0.000). Conclusion Estradiol has no obvious effect on intraocular pressure in rats with high intraocular pressure, but it can increase retinal thickness, inhibit apoptosis and autophagy in retina.
