摘要
目的:探讨"过敏煎"联合咪唑斯汀对亚急性接触性湿疹小鼠模型的影响。方法:选取健康清洁级ICR系小鼠40只,适应性喂养1周后随机分为空白对照组、2,4-二硝基氯苯(DNCB)模型组、过敏煎组、咪唑斯汀组、过敏煎+咪唑斯汀组,除空白对照组外,将7%、1%的DNCB-丙酮溶液分别涂抹在其余各组小鼠右耳部及背部脱毛处皮肤,制作小鼠亚急性接触性湿疹模型。各组均在实验造模第2天开始按相应剂量[20m L/kg(体质量)]灌胃给药,1次/d,连续给药10天,空白对照组及DNCB模型组给予等体积去离子水。观察小鼠右耳肿胀程度、两耳重量差、厚度差,观察各组小鼠右耳中部组织真皮炎症细胞数及10分钟舔体反应次数。采用ELISA法检测各组小鼠血清中白细胞介素4(IL-4)、干扰素γ(IFN-γ)及白细胞介素2(IL-2)的含量。结果:过敏煎联合咪唑斯汀对DNCB致小鼠耳廓肿胀有明显抑制作用,对4-AP诱发小鼠舔体反应的皮肤瘙痒有显著抑制作用,两药联合可通过直接升高湿疹小鼠血清中IFN-γ和IL-2,从而间接降低IL-4表达,改善湿疹情况。结论:过敏煎联合咪唑斯汀对亚急性湿疹模型小鼠耳廓肿胀及皮肤瘙痒有明显抑制作用,其作用机理与抗炎、止痒及调节细胞因子分泌紊乱有关。
Objective: To discuss the effects of "GuoMin decoction" combined with mixolastine on the mice model suffering subacute eczema. Methods: Forty mice accepted adaptive feeding for one week, randomized into blank control group, DNCB model group, GuoMin decoction group, mixolastine group, the combination group of GuoMin decoction and mixolastine, except blank control group, 7% and 1% DNCB-acetone solution were applied to the bare skin of right ear and the back of the mice in the other groups respectively to prepare the mice models with subacute exzema. Different groups were medicated since the second day after preparing the models by lavaging in the corresponding dosage, 20 m L/kg, once each day, for ten consecutive days, blank control group and DNCB model group were given the equal volume of deionized water. To observe the swelling degrees of right ear, the weight difference between two ears, thickness differences of the mice, to observe the number of pathological dermis inflammatory cells in the middle region of right ear and licking response times within ten minutes. To detect the contents of IL-2, IFN-γ and IL-4 in the serum of the mice in different groups by ELISA method. Results: GuoMin decoction combined with mixolastine showed the inhibitory effects on the mice model with DNCB-induced auricle swelling and inflammation apparently, and 4-AP-induced licking response in the mice model with skin itching, the combination of two drugs could directly raise the levels of IL-2 and IFN-γ of the mice with eczema, thus to lower the expression of IL-4 indirectly and improve the conditions of eczema. Conclusion: GuoMin decoction and mixolastine possess the inhibitory effects on the mice suffering subacute eczema, and the mechanism is related to anti-inflammatory effects, relieving the itching and regulating cytokine secretion disorder.
