摘要
目的探讨miR-21,miR-375及其相关靶基因在喉癌中的表达及微小RNA与靶基因的相关性。方法实时荧光定量PCR方法检测喉癌及癌旁组织中miR-21和miR-375及其靶基因BTG2与PDK1的转录水平表达,Western blot法检测BTG2与PDK1的翻译水平表达。结果在喉癌组织中miR-21、PDK1高表达,miR-375、BTG2低表达,miR-21与BTG2、miR-375与PDK1均呈负相关。结论在喉癌中miR-21可能是癌基因,对其靶基因BTG2起负调控作用;miR-375可能是抑癌基因,对其靶基因PDK1起负调控作用。
Objective To investigate the expression of miR-21,miR-375 in the laryngocarcinoma tissues,and demonstrate the relations of microRNAs and their target genes.Methods The transcriptional expression levels of miR-21 and miR-375 were detected by the real-time PCR,the translational expression levels of BTG2 and PDK1 were detected by Western blot.Results Compared with adjacent tissues,the expression level of miR-21 was up-regulated in the cancer tissues,which had a negative relationship with its target gene BTG2.Moreover,the expression level of miR-375 was down-regulated in the cancer tissues,which had the same negative relationship with its target gene PDK1.Conclusion In the laryngocarcinoma,the miR-21 might act as a cancer-promoting genes to suppress the BTG2 protein product.On the contrary,the miR-375 might play a cancer-inhibiting role in the suppression of protein expression of PDK1.
