摘要
免疫学实验课是免疫学教学的重要组成部分,对于培养学生的实践能力和科研思维具有重要作用。小鼠腹腔巨噬细胞免疫细胞化学染色试验是学生需掌握的重要的免疫学实验技术之一,然而应用商品化的单克隆抗体进行检测有阳性率低、检测结果不稳定及成本高等问题。因此,我们尝试了通过自主制备多克隆抗体检测巨噬细胞的方法应用于实践教学中,取得了良好的效果。研究表明采用自制多克隆抗体能明显增加细胞阳性染色比例,且染色更加明显,利于结果观察和判断,当抗体稀释至2 000倍后仍能检测到巨噬细胞。并且抗体制备简单、来源方便,成本降低、易于推广,为在免疫学实践课中开展巨噬细胞的检测提供了必要的手段。
As a supplement to the theory course,immunology practice teaching plays an important role in cultivating students′practical ability and scientific research thinking.Immunocytochemical staining of mouse peritoneal macrophages is one of the important immunological experimental techniques,and it is an important technique that students need to master.By summarizing the problems in the past teaching,we try to explore the methods of preparing polyclonal antibodies for detecting macrophages,and apply them to practical teaching.It has achieved good results.The research has showed that the positive ratio of staining for mouse peritoneal macrophages has been markedly increased that make the observation and assessment of the results efficiently by using prepared polyclonal antibodies.In addition,this kind of antibodies can be simple and convenient prepared,making the cost reduction and easy for application,that could provide the useful tool for macrophages study in immunological experiment class.
