摘要
2018年3月17日,贵州省某梅花鹿场的梅花鹿(Cervus nippon)出现关节肿大、下颌肿大、睾丸单侧肿胀,还伴有严重的跛行等症状。采集4份该发病梅花鹿的血样送检。检测结果表明,通过虎红平板凝集试验2份血样检测出了布氏杆菌感染,4份血样经RT-PCR均扩增出了JEV NS1部分基因的特异性条带,PCR产物测序结果分析显示,4份梅花鹿血样的JEV NS1基因(GZ-015、GZ-019、GZ-022、GZ-039)间的核苷酸同源性分别为99. 9%、99. 3%、99. 4%,与强毒株SA-14、国内外的疫苗株及四川、吉林等地的JEV分离株之间核苷酸同源性为99%以上,GZ-015、GZ-019、GZ-022、GZ-039与强毒株SA-14、弱毒株、国内外的疫苗株及四川、吉林等地的JEV分离株处于同一个较大的遗传进化分支。
On 17 March 2018,sika deer(Cervus nippon)in a sika deer farm in Guizhou Province had developed symptoms,including swollen joints,swollen jaws,unilateral swelling of the testicles,accompanied by severe lameness.The farm staff collected 4 blood samples from the affected sika deer for laboratory analyses.The test results showed that Brucella infection was detected in 2 samples by tiger red plate agglutination test,and 4 blood samples were amplified at JEV NS1 specific bands by RT-PCR.The sequencing analysis of PCR products showed that the nucleotide homologies among the JEV NS1 genes(GZ-015,GZ-019,GZ-022,and GZ-039)of the blood samples from four sika deer were 99.9%,99.3%,and 99.4%,respectively,and the virulent strain SA-14,domestic and foreign vaccine strains and the nucleotide homology between JEV isolates in Sichuan and Jilin were greater than 99%.GZ-015,GZ-019,GZ-022,GZ-039 and virulent strains SA-14,attenuated strains,vaccine strains at home and abroad,and JEV isolates from Sichuan and Jilin are in the same branch of genetic evolution.
