消瘀降脂胶囊预防给药对高脂血症模型兔血脂及相关蛋白酶含量的影响 被引量:5

Effects of Preventive Administration of Xiaoyu Jiangzhi Capsule(消瘀降脂胶囊) on Blood Lipid and Related Protease Content in Hyperlipidemia Model Rabbits

摘要 目的探讨消瘀降脂胶囊预防高脂血症的效果及可能作用机制。方法雄性新西兰兔40只随机分为正常对照组、模型对照组、阿托伐他汀组、消瘀降脂胶囊组以及联合给药组,每组8只。正常对照组每日予以基础饲料喂养,其余各组以高脂饲料喂养。从第2周开始,正常对照组与模型对照组每日予以30 ml生理盐水灌胃,阿托伐他汀组给予2 mg/(kg·d)阿托伐他汀片灌胃,消瘀降脂胶囊组给予100 mg/(kg·d)消瘀降脂胶囊灌胃,联合给药组同时给予2 mg/(kg·d)的阿托伐他汀片以及100(kg·d) mg/kg的消瘀降脂胶囊灌胃,连续给药10周。检测各组实验兔血脂及血清中低密度脂蛋白受体(LDL-R)、载脂蛋白B100(Apo-B100)、脂蛋白脂酶(LPL)、前蛋白转化酶枯草溶菌素9 (PCSK9)含量,肝脏中3-羟基-3-甲基戊二酰辅酶A还原酶(HMGCR)、过氧化物酶增殖物活化受体γ(PPAR-γ)、固醇调节元件结合蛋白2(SREBP2)、微粒体三酰甘油转移蛋白(MTP)以及小肠胆固醇转运蛋白(NPC1L1)含量。结果与正常对照组比较,模型对照组兔血脂水平升高,血清PCSK9、Apo-B100含量升高,LDL-R、LPL含量降低,肝组织中HMGCR、PPAR-γ、MTP及小肠组织中NPC1L1升高,肝组织SREBP2含量降低(P <0. 05或P <0. 01)。与模型对照组比较,各给药组兔血脂水平均不同程度降低,并且以联合给药组降低更明显(P <0. 05或P <0. 01);各给药组兔血清、肝脏组织、小肠组织中各相关蛋白酶含量有不同程度改善,且以联合给药组改善最明显(P <0. 05或P <0. 01)。结论消瘀降脂胶囊可能通过对脂质代谢相关酶的调控,抑制小肠中的胆固醇吸收以及肝脏细胞内胆固醇合成,并加强血液中甘油三酯水解等作用,从而发挥对高脂血症的预防作用。 Objective To explore the effects and possible mechanism of Xiaoyu Jiangzhi Capsule(消瘀降脂胶囊)in preventing hyperlipidemia.Methods A total of 40 male New Zealand rabbits were randomly divided into normal control group,model control group,atorvastatin group,Xiaoyu Jiangzhi Capsule group and combined drug group,with 8 rats in each group.The normal control group was fed with basic diet daily,and the other groups were fed with high fat diet.From the 2 nd week,the normal control group and the model control group were given 30 ml of normal saline daily,and the atorvastatin group was given 2 mg/(kg·d)atorvastatin tablets by gavage,and the Xiaoyu Jiangzhi Capsule group was intragastrically administered with 100 mg/(kg·d)Xiaoyu Jiangzhi Capsule.The combined administration group was given 2 mg/(kg·d)atorvastatin tablets and 100 mg/(kg·d)Xiaoyu Jiangzhi Capsules by gavage for 10 weeks.Blood lipids,serum low density lipoprotein receptor(LDL-R),apolipoprotein B100(Apo-B100),lipoprotein lipase(LPL),proprotein convertase subtilisin/kexin type 9(PCSK9)content,liver content of 3-hydroxy-3-methylglutaryl coenzyme A reductase(HMGCR),peroxisome proliferator-activated receptorγ(PPAR-γ),sterol regulatory element binding protein 2(SREBP2),microsomal triacylglycerol transfer protein(MTP)and Niemann-Pick C1-Like 1(NPC1 L1)were detected in each group of experimental rabbits.Results Compared with the normal control group,the rabbits in the model control group had elevated blood lipid levels,elevated serum PCSK9 and Apo-B100 levels,decreased LDL-R and LPL levels,and HMGCR,PPAR-γ,MTP in the liver tissue and NPC1 L1 in the small intestine were increased,while liver tissue SREBP2 content decreased(P<0.05 or P<0.01).Compared with the model control group,the blood lipid levels of the rabbits in each administration group were decreased to different degrees,and the decrease was more significant in the combined drug group(P<0.05 or P<0.01).The serum,liver tissue and small intestine tissues of the rabbits in each drug group were impro
