大鼠肝部分切除术后肝细胞生长因子、转化生长因子-β1水平变化及脾动脉结扎对大鼠肝再生的影响 被引量:8

摘要 目的测定肝部分切除术后大鼠血清肝细胞生长因子(HGF)、转化生长因子-β1(TGF-β1)水平,探讨脾动脉结扎对大鼠肝再生的影响。方法将90只雄性SD大鼠随机分为3组:假手术组(Sham组):单纯开腹并游离脾动静脉及肝周韧带;对照组(Ph组):建立经典的70%肝部分切除模型;实验组(Ph+Sal组):在Ph组基础上,行脾动脉结扎。分别于术后24、48、72 h,取出大鼠肝脏并计算肝体比、使用免疫组织化学检测肝组织中HGF、TGF-β1的表达,采用酶联免疫吸附试验(ELISA)检测血清中HGF、TGF-β1的水平。结果Sham组大鼠术后24、48、72 h肝体比变化不明显,差异无统计学意义(P>0.05)、血清中TGF-β1水平差异无统计学意义(P>0.05),3组大鼠术后24、48、72 h血清中HGF水平差异无统计学意义(P>0.05),Sham组术后同时间[(35.28±7.02)、(35.20±9.01)、(35.15±7.10) pg/ml]与Ph组[(456.16±15.90)、(426.22±13.90)、(418.55±12.21) pg/ml]、Ph+Sal组[(470.15±14.60)、(438.12±12.05)、(410.15±11.20) pg/ml]血清HGF水平差异有统计学意义(P<0.05)、Ph和Ph+Sal血清中TGF-β1水平随时间逐渐增高,术后同时间血清中TGF-β1水平均高于Sham组(P<0.05),Ph组大鼠术后同时间肝体比显著小于Ph+Sal组,差异有统计学意义(P<0.05);Ph组大鼠术后24、48、72 h血清中TGF-β1水平均高于Ph+Sal组大鼠(P<0.05)。结论脾动脉结扎可降低门脉压力,同时可能通过升高HGF水平、降低TGF-β1水平,促进肝再生,表明脾动脉结扎可以促进肝再生。 Objective To investigate the effects of splenic artery ligation on rat liver regeneration by determining the levels of hepatocyte growth factor (HGF) and transforming growth factor-β1 (TGF-β1) after partial hepatectomy. Methods Ninety male SD rats were randomly divided into 3 groups: Sham operation group (Sham group)(opening the abdomen and dissociating the splenic artery and vein and the perihepatic ligament);control group (Ph group)(the classic 70% partial liver resection model established). In the experimental group (Ph+ Sal group), splenic artery ligation was performed on the basis of Ph group. The rat liver was taken out at 24, 48 and 72 h after the operation, and the liver body ratio was calculated. The expression of HGF and TGF-1 in liver tissues was detected by immunohistochemistry. The serum levels of HGF and TGF-1 were determined by enzyme linked immunosorbent assay (ELISA). Results Sham group of rats after 24, 48, 72 h liver than change is not obvious, no statistical difference (P>0.05), the difference of serum level of TGF-beta 1 no statistical significance (P>0.05), three groups of rats after 24, 48, 72 h in serum HGF level no significant statistical difference (P>0.05), Sham group began with time [(35.28±7.02),(35.20±9.01),(35.15±7.10) pg/ml] and Ph [(456.16±15.90),(426.22±13.90),(418.55±12.21) pg/ml], Ph+ Sal group [(470.15±14.60),(438.12±12.05),(410.15±11.20) pg/ml] serum HGF level difference is apparent, there was significant statistical significance (P<0.05), the level of TGF-aspiration 1 in serum of Ph and Ph+ Sal increased gradually over time, and the level of TGF-aspiration 1 in serum at the same postoperative time was higher than that of Sham group (P<0.05). The liver body ratio of rats at the same postoperative time was significantly lower than that of Ph+ Sal group (P<0.05). The serum TGF-proge1 level in 24, 48 and 72 h postoperatively was higher in rats of Ph+ Sal group (P<0.05).
