基于成分含量与抗肿瘤抗氧化作用相关性分析的不同产地薏苡仁质量评价研究 被引量:4

Quality evaluation of coix seed based on correlation analysis of the contents of active ingredients with anti-tumor and antioxidant activity

摘要 目的探明薏苡仁成分含量与对肝癌细胞的增殖抑制作用以及抗氧化作用的相关性,评价不同产地薏苡仁质量。方法分别采用HPLC-ELSD法和气相色谱法测定不同产地薏苡仁中甘油三油酸酯与1, 2-油酸-3-亚油酸甘油三酯、油酸与亚油酸的含量;采用清除1, 1-二苯基苦基苯肼法(DPPH)和四甲基偶氮唑盐比色法(MTT)评价不同产地薏苡仁体外抗肿瘤活性以及抗氧化能力,并对成分含量和活性结果进行多元线性回归分析。结果经多元线性回归法分析,由SPSS 21.0建立以下方程:Y_1=1.636 x_1^(**)+1.331 x_2^(**)+1.365 x_3+0.225 x_4-24.853;Y_2=1.878 x_1^(**)-0.061 x_2-4.962 x_3~*+9.918 x_4~*+35.007(~*P<0.05,^(**)P<0.01)。甘油三油酸酯及1, 2-油酸-3-亚油酸甘油三酯总含量与抗肿瘤作用呈正相关;甘油三油酸酯及亚油酸总含量与抗氧化作用呈正相关。结论单一成分含量高低与薏苡仁活性大小并不吻合,采用与活性为正相关的多指标成分的总含量作为薏苡仁质量评价方法,较之单一甘油三油酸酯为指标的评价方法更为科学合理。15个产地薏苡仁质量差异较大,其中以辽宁葫芦岛的薏苡仁质量较优。 Objective To investigate the correlation of contents of active ingredients between anti-proliferative effect against human hepatoma HepG2 cell and antioxidant capacity,and to evaluate the quality of coix seed from fifteen places.Methods HPLC-ELSD and GC were employed to determine contents of triglyceride,1,2-oleic acid-3-linoleic acid triglyceride,oleic acid and linoleic acid in coix seed,respectively.In addition,MTT assay and DPPH assay were performed to explore anti-tumor and antioxidant capacity of coix seed from various origins.Furthermore,multiple linear regression analysis was conducted on the contents of active ingredients and the results of pharmacodynamic activity.Results By multivariate linear regression analysis,the following equations were established by SPSS 21.0:Y1=1.636 x1^**+ 1.331 x2^**+1.365 x3 + 0.225 x4-24.853;Y2=1.878 x1^**-0.061 x2-4.962 x3^*+ 9.918 x4^*+ 35.007.And the total contents of triglyceride and 1,2-oleic acid-3-linoleic acid triglyceride in coix seed were positively correlated with the anti-tumor effect.The total contents of triglyceride and linoleic acid were positively correlated with the antioxidant capacity.Conclusion The content of single component was not coincide with the pharmacodynamic activity of coix seed,and the quality evaluation method that referred the total contents of multi-index components positively correlated with pharmacodynamic activity,which was more scientific and reasonable than referred only single index of triglyceride.The quality of coix seed in 15 places is quite differently,among them,the quality in Liaoning Huludao is the best.
