聚苯乙烯磺酸镧Beagle犬体内药动学研究

摘要 目的建立电感耦合等离子质谱仪(ICP-MS)测定血浆中镧的方法,研究聚苯乙烯磺酸镧在犬体内的药动学特征。方法 18只Beagle犬随机分成高、中、低剂量组,禁食12 h后,于早晨分别口服给予聚苯乙烯磺酸镧1.5,0.75,0.375 g/kg,采用电感耦合等离子体质谱仪(ICP-MS)测定血浆中的镧含量。聚苯乙烯磺酸镧血药浓度-时间数据经DAS1.0处理得药动学参数。结果犬口服单次给予1.5,0.7,0.375 g/kg聚苯乙烯磺酸镧,C_(max)分别为149.6±50.6,71.4±12.3,40.8±7.2μg/L,T_(max)分别为3.4±0.7,3.3±1.0,3.6±0.5 h,t1/2Z分别为9.0±4.2,5.7±5.1,9.6±11.7 h,AUC_(0-24 h)分别为817.6±114.4,455.9±96.7,296.9±31.3μg/(L·h),MRT_(0-24h)分别为7.4±0.8,8.1±0.5,8.3±0.4 h。结论利用ICP-MS可准确定量血浆样本中镧的含量,获得聚苯乙烯磺酸镧在犬体内的药动学参数。 Objective To establish an ICP-MS method for the determination of polystyrene sulfonic lanthanum in plasma and study the pharmacokinetic characteristics of polystyrene sulfonic lanthanum in dogs.Methods 18 Beagle dogs were randomly divided into high-dose group,middle-dose group and low-dose group.After fasting for 12 hours,1.5,0.75 and 0.375 g/kg of polystyrene sulfonic lanthanum were orally administered in the morning,and the lanthanum concentration in plasma was measured by ICP-MS.The plasma concentration-time data were processed by DAS 1.0 to obtain the pharmacokinetic parameters of polystyrene sulfonic lanthanum.Results Cmax of the dogs given a single oral dose of 1.5,0.75 and 0.375 g/kg of polystyrene sulfonic lanthanum were 149.6±50.6,71.4±12.3 and 40.8±7.2μg/L;Tmax were 3.4±0.7,3.3±1.0 and 3.6±0.5 h;t1/2Z were 9.0±4.2,5.7±5.1 and 9.6±11.7 h;AUC0-24 h were 817.6±114.4,455.9±96.7 and 296.9±31.3μg/(L·h);MRT0-24 h were 7.4±0.8,8.1±0.5 and 8.3±0.4 h.Conclusion The ICP-MS can accurately quantify the content of lanthanum in biological samples and obtain the pharmacokinetic parameters of polystyrene sulfonic lanthanum in dogs.
