电针对阿尔茨海默病大鼠齿状回星形胶质细胞和白细胞介素-10表达及其共定位的影响 被引量:14

Effects of electroacupuncture on the expression and co-localization of hippocampal dentate astrocytes and IL-10 in AD rats

摘要 目的观察电针对阿尔茨海默病(AD)大鼠海马齿状回星形胶质细胞(AC)和白细胞介素(IL)-10表达及其共定位的影响。方法 40只雄性Wistar大鼠随机等分成对照组、假手术组、模型组和治疗组。采用双侧海马注射Aβ1~42建立AD模型。对照组、假手术组和模型组不予任何治疗;治疗组选取百会、肾俞,电针20 min,1次/d,6 d为1个疗程,2个疗程间休息1 d。采用免疫组化法和免疫双标法检测相关指标。结果模型组和治疗组胶质纤维酸性蛋白(GFAP)阳性表达均显著升高(P<0. 01),但两组间无显著性差异(P>0. 05),模型组AC胞体变大,突起增多增粗,胞核空化,染色深。模型组IL-10阳性表达显著降低(P<0. 01),治疗组IL-10阳性表达显著增强(P<0. 01)。假手术组IL-10与GFAP共定位呈金黄色,模型组IL-10主要定位于神经元细胞核,治疗组IL-10表达显著增强,不仅表达在神经元细胞核,也表达在神经元纤维。结论电针对AD的调节作用不限于抗炎作用,尚可促进AC对神经元的抗炎保护作用。 Objective To observe the effects of electroacupuncture on the expression and co-localization of hippocampal dentate astrocytes and IL-10 in AD rats.Methods Forty healthy male Wistar rats were randomly divided into normal control,sham operation,model and EA treatment groups,each group had 10 rats.Both model and EA groups were bilaterally injected into the hippocampal region with Aβ1-42;normal control,sham operation group and model groups were treated without any treatment.EA group was selected"Bai hui"(GV20)and bilateral"Shen shu"(BL23)acupoint.An individual EA session was administered daily for 20 min,6 days,and 1 day rest for a course of 6 days,totally for 2 courses.The related results were detected by immunohistochemistry and double-labelling immunofluorescence.Results Compared with that of control group,GFAP positive expressions were increased both in model and treatment groups(P<0.01),but there was no significant difference between the two latter groups(P>0.05).The positive expression of IL-10 was significantly decreased in model group(P<0.01)and significantly increased in treatment group(P<0.01).IL-10 and GFAP were co-located position in sham group,but in model group,IL-10 was mainly located in the nucleus.Conclusions The regulatory effects of electroacupuncture on AD astrocytes are not limited to anti-inflammatory effects.
