摘要 目的研究miR-155对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)的影响及其与细胞因子白细胞介素-35(IL-35)的相关性。方法选取稳定期COPD患者30例(COPD稳定组),COPD急性加重期患者30例(AECOPD组),重度吸烟组30例,健康对照组30例。各组受试者分别检测肺功能及血气分析。抽取各组外周血,RT-PCR检测外周血单个核细胞(PBMC)中miRNA-155的表达水平,ELISA方法检测细胞因子IL-35的表达水平。分析miRNA-155与血气指标、肺功能、IL-35之间的相关性。结果 AECOPD组miR-155表达量最高,COPD稳定组表达次之,重度吸烟组表达较少,健康对照组表达最少。IL-35的表达结果与其恰好相反。对miR-155与PO_2、FEV1%、IL-35的水平进行相关性分析,结果显示miR-155与上述指标均存在负相关关系(r=-0.739 7、-0.706 2、-0.652 2),且具有显著统计学意义(P<0.01)。结论 miR-155参与了COPD的发病过程,是导致肺损伤的重要物质,对COPD产生负性效应。 Objective To study the effect of miR-155 on chronic obstructive pulmonary disease(COPD) and its correlation with cytokine IL-35. Methods 30 patients with stable COPD from the respiratory department of our hospital from June 2017 to June 2018, 30 patients with acute exacerbation of COPD(AECOPD group), 30 patients with severe smoking, and 30 healthy controls were enrolled. Each participants were tested with lung function and blood gas analysis. The peripheral blood of each group was extracted, the expression level of miRNA-155 in PBMC of mononuclear cells was detected by RT-PCR, and the expression level of cytokine IL-35 was detected by ELISA. The correlations between miRNA-155 and blood gas index, lung function, and IL-35 were analyzed. Results The expression of miR-155 was the highest in the AECOPD group, and followed by the COPD stable group, severe smoking group and control group. The expression of IL-35 was exactly the opposite. There were significant negative correlations between miR-155 and PO2, FEV1% and IL-35 levels(r=0.739 7, 0.706 2 and 0.652 2, respectively;P<0.01). Conclusion MiR-155 is involved in the pathogenesis of COPD and is an important substance leading to lung injury, which has a negative effect on chronic obstructive pulmonary disease.
