摘要
目的:运用HPLC法同时测定美洲大蠊提取物中9种核苷类成分。方法:取美洲大蠊提取物冻干粉以流动相(2.5%甲醇)为溶剂进行溶解,配制成0.02 mg﹒m L^(-1)的供试品溶液,使用反相C18烷基硅胶柱(250 mm×460 mm,5μm),以纯水:甲醇=97.5:2.5为流动相,体积流量1.0 mL﹒min^(-1),检测波长254 nm;柱温32℃,进样量为10μL为检测条件,对供试品溶液进行含量测定。结果:通过对3批美洲大蠊提取物冻干粉进行测定,结果显示胞嘧啶、尿嘧啶、胞苷、次黄嘌呤、尿苷、胸腺嘧啶、腺嘌呤、肌苷、鸟苷的平均含量分别为6.195 mg﹒g^(-1)、0.732 mg﹒g^(-1)、1.310 mg﹒g^(-1)、2.465 mg﹒g^(-1)、1.418 mg﹒g^(-1)、0.445 mg﹒g^(-1)、2.846 mg﹒g^(-1)、2.223 mg﹒g^(-1)、1.742 mg﹒g^(-1),平均总含量为19.376 mg﹒g^(-1)。结论:所建立的HPLC方法在对同时测定美洲大蠊提取物中9种核苷类成分的测定中具简便、可靠、稳定、可行的特点,可用于康复新系列产品中间体的质量控制。
Objective: To simultaneously detect 9 nucleosides in Periplaneta Americana L.extract by HPLC. Method: The freeze-dried Periplaneta Americana L.extract were dissolved in mobile phase(2.5% methanol of water solution) to get the test solution of 0.02 mg﹒m L^-1. The reverse phase C18 alkyl silica gel column(250 mm × 460 mm, 5 μm) was applied in HPLC determination with water and methanol mixture( 97.5:2.5) as the mobilephase at the flow rate of 1.0 mL﹒min^-1. Detection wavelength was set at 254 nm, and column temperature was 32 ℃.10 μL sample was injected for analysis. Result: 3 batch of Periplaneta Americana L.extract freeze-dried powder was detected.The results showed that the average content of cytosine, uracil, cytidine, hypoxanthine, uridine, thymidine, adenine, inosine and guanosine were 6.195 mg﹒g^-1, 0.732 mg﹒g^-1, 1.310 mg﹒g^-1, 2.465 mg﹒g^-1, 1.418 mg﹒g^-1, 0.445 mg﹒g^-1, 2.846 mg﹒g^-1, 2.223 mg﹒g^-1,and 1.742 mg﹒g^-1.The average total content of nucleosides was 19.376 mg﹒g^-1. Conclusion: HPLC method is simple, reliable, stable and feasible for the simultaneous determination of the 9 kinds of nucleosides of Periplaneta Americana L.extract. It can be used as a quality control method for intermediates of Kangfuxin products.
