摘要
目的探讨应用烟熏联合气管注入脂多糖复合法制备慢性阻塞性肺疾病(COPD)大鼠模型的注意事项。方法选取7周龄Wistar雄性大鼠60只,随机分成模拟手术组(10只,A组)和造模组(50只,B组)。观察2组大鼠体征表现、便秘情况、死亡情况,以及病理检查情况。结果 B组大鼠尿量较A组明显减少,体重较A组下降。B组大鼠死亡18只(36.00%),便秘8只(16.00%),29份(90.62%)气管、肺组织样本符合COPD病理改变。A组死亡1只(10.00%),无明显便秘,9份(100.00%)气管、肺组织样本病理检查结果正常。结论烟熏联合气管注入脂多糖复合造模法可致大鼠呈COPD病理改变,但需注意造模过程中大鼠状态、烟雾含量、温度、湿度等细节控制。
Objective To explore the precautions for the preparation of chronic obstructive pulmonary disease(COPD) rat model by smoke inhalation combined with intratracheal injection of lipopolysaccharide(LPS). Methods 60 seven-week-old Wistar male rats were randomly divided into simulated operation group(10 rats,group A) and model group(50 rats,group B).The signs,constipation,death and pathological examination of the two groups of rats were observed. Results The urine volume and body weight of rats in the group B were significantly lower than those in the group A.In the group B,18 rats(36.00%) died and 8 rats(16.00%) suffered from constipation.29 trachea and lung tissue samples(90.62%) were consistent with COPD pathological changes.In the group A,1 rat died(10.00%) without obvious constipation,and the pathological results of 9 trachea and lung tissues samples(100.00%) were normal. Conclusion Smoke inhalation combined with LPS injection into trachea can induce pathological changes of COPD in rats,but attention should be paid to the detailed control of rat state,smoke content,temperature and humidity.
