摘要
目的探讨蛋白激酶C对早期运动预处理心肌肌膜ATP敏感钾通道SUR2A亚基表达的影响。方法选取健康雄性SD大鼠32只,在动物实验跑台上以15 m/min的速度进行为期5 d的适应性训练,按照随机数字表法分为对照组、力竭运动组(EE组)、早期运动预处理+力竭运动组(EEP+EE组)和蛋白激酶C阻断剂+早期运动预处理+力竭运动组(CHE+EEP+EE组),每组各8只。对照组不进行任何跑台运动训练。EE组以35 m/min的速度进行力竭运动训练,建立大鼠运动性心肌损伤模型。EEP+EE组先以28~30 m/min的速度运动10 min,休息10 min,重复4次,建立EEP动物模型,30 min后以35 m/min的速度运动训练直至力竭。CHE+EEP+EE组在运动前腹腔注射5 mg/kg的白屈菜赤碱,其余运动方法同EEP+EE组。检测心肌SUR2A的表达情况。结果与对照组比较,EE组SUR2A原位杂交阳性反应面积和积分光密度值增强,EE组、CHE+EEP+EE组SUR2A mRNA表达水平与SUR2A表达水平均升高,差异具有统计学意义(P<0.05);与EE组比较,EEP+EE组SUR2A原位杂交阳性反应面积和积分光密度降低,SUR2A mRNA与SUR2A表达水平均下降,差异具有统计学差异(P<0.05);与EEP+EE组比较,CHE+EEP+EE组SUR2A原位杂交阳性反应面积和积分光密度增强,SUR2A mRNA与SUR2A表达水平均升高,差异具有统计学差异(P<0.05)。结论蛋白激酶C通过调控SUR2A的表达以介导早期运动预处理保护心肌。
Objective To investigate the effect of protein kinase C (PKC) on the expression of SUR2A subunit of ATP-sensitive potassium channel in myocardium during early exercise preconditioning. Methods Thirty-two healthy male SD rats were selected and trained at a speed of 15 m/min for 5 days on the experimental treadmill. They were divided into control group, exhaustive exercise group (EE group), early exercise pretreatment + exhaustive exercise group (EEP + EE group) and protein kinase C (PKC) blocker + early exercise pretreatment + exhaustive exercise group (CHE + EEP + EE group), with 8 rats in each group. The control group did not undergo any treadmill training. In EE group, exhaustive exercise training was conducted at a speed of 35 m/min to establish a rat model of exercise-induced myocardial injury. In EEP + EE group, the EEP animal model was established by 10-min exercise at a speed of 28-30 m/min , and 10-min rest , which was repeated four times. After 30 min, the EEP animal model was trained at a speed of 35 m/min until exhaustion. CHE + EEP + EE group received intraperitoneal injection of 5 mg/kg chelerythrine before exercise. The remaining the exercise methods were identical with those of EEP + EE group. The expression of SUR2A in myocardium was detected. Results Compared with the control group, the positive reaction area and integral optical density of SUR2A in situ hybridization in EE group were increased, and the expression levels of SUR2A mRNA and SUR2A in EE group and CHE+EEP+EE group were increased, suggesting significant difference ( P <0.05). Compared with EE group, the positive reaction area and integral optical density of SUR2A in situ hybridization in EEP+EE group were decreased, and the expression levels of SUR2A mRNA and SUR2A were both increased, indicating significant differences ( P <0.05). The positive reaction area and integral optical density of SUR2A mRNA in CHE+EEP+EE group were increased, and the expression levels of SUR2A mRNA and SUR2A were increased in CHE+EEP+EE group compared with E
