七氟醚对新生大鼠海马神经元NMDA受体1亚基和2B亚基mRNA的影响 被引量:2

摘要 目的观察七氟醚对新生大鼠海马组织NMDA受体1亚基和2B亚基mRNA的表达水平及大鼠在学习、记忆能力方面的影响。方法将48只出生后7 d的SD乳鼠,体质量10~15 g,雌雄不限,随机分为两组,即七氟醚吸入组(S组):吸入2.3%七氟醚4 h;对照组(D组):吸入氧气4 h,各24只。麻醉结束后,立即随机取S组8只(S1组)、D组8只(D1组)断头取脑,分离海马组织,进行实时荧光定量PCR检测NMDA受体l亚基和2B亚基mRNA的表达情况。剩余的S组和D组乳鼠各16只,在空气条件下饲养7 d后,随机各取8只分为S2组和D2组,取大脑海马组织重复实时荧光定量PCR检测NMDA受体l亚基和2B亚基mRNA的表达情况。最后存活的乳鼠各8只,即S3组和D3组,进行跳台试验。结果与对照组(D1组和D2组)比较,吸入七氟醚的实验组(S1组和S2组),乳鼠的大脑海马组织的NMDA受体1亚基、2B亚基mRNA表达降低,与对照组(D3组)比较,吸入七氟醚的实验组(S3组)乳鼠学习成绩、记忆成绩显著降低,差异均有统计学意义(P<0.05)。结论持续暴露于七氟醚中可降低未成熟SD大鼠学习、记忆能力,其原因可能与NMDA受体l亚基、2B亚基mRNA的表达降低有关。 Objective To observe the mRNA expression level of subunit 1 and 2B of NMDA receptor in the hippocampus of Sprague Dawley (SD) model rats,the effect of Sevoflurane on the learning and memory abilities. Methods The neonatal SD rats of weighting 10~15 g, 7 days after birth and the number of 48, sex not limited, were randomly divided into two groups, 24 of Sevoflurane inhalation group (S group) which was inhaled 2.3% Sevoflurane for 4 hours, 24 of control group (group D) which was inhaled oxygen for 4 hours. After end of anesthesia, immediately randomly took out 8 rats from group S (S1 group), 8 rats from group D (D1 group).And then broke out the brains, separated hippocampal tissue. The mRNA expression levels of subunit 1 and 2B of NMDA receptor were determined by the real-time quantitative PCR. The remaining S and D group with 16 rats in each, after 7 days feeding in the air condition, randomly selected from S and D group with 8 rats for each were divided into S2 group and D2 group. The mRNA expression levels of subunit 1 and 2B of NMDA receptor were determined by the real-time quantitative PCR again. Finally, the survival rats of 16 from S and D group, namely S3 group and D3 group, to the step-down test. Results Compared with the control group (D1 group and D2 group), the rat brain hippocampal mRNA expression level of submit 1 decreased and the mRNA expression level of subunit 2B also decreased in inhalation of Sevoflurane test groups (S1group and S2 group)( P <0.05, P <0.05). Compared with the control group (D3), inhalation of Sevoflurane in the test group (S3 group) rats learning were significantly decreased ( P <0.05), memories performance were significantly lower ( P <0.05). Conclusion The exposure to Sevoflurane can reduce learning and memory abilities of SD rats by decreasing the mRNA expression level of subunit 1 and 2B of NMDA receptor.
