摘要 目的采用傅里叶变换红外(Fourier transform infrared,FTIR)显微光谱技术分析烧死者支气管上皮生化成分变化,为烧死案件鉴定提供辅助手段。方法采用10例烧死和20例非烧死案例,使用与红外显微镜联合的傅里叶变换红外光谱仪检测肺支气管上皮组织。对得到的红外光谱进行主成分分析和偏最小二乘判别分析。结果与对照组相比,烧死组支气管上皮细胞含有更多的β-转角和β-折叠以及更少的α-螺旋蛋白二级结构。构建的PLS-DA分类模型用于识别烧死,表现出优异的分类能力,准确率达到100%。结论红外光谱结合化学计量学方法可作为烧死死后诊断的补充手段。 Objective Fourier transform infrared(FTIR)microscopic spectroscopy was used to analyze the alternations of biochemical components in the bronchial epithelium of fatal burn cases in order to provide a supplementary method for the post-mortem diagnosis of fatal burn.Method 10 cases of fatal burns and 20 control cases were used in this study.The spectra of lung bronchial epitheliums were measured by FTIR microscopic spectroscopy.The principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA)were performed to analyze the obtained infrared spectra.Results The bronchial epitheliums of fatal burn group contained moreβ-turn andβ-sheet and lessα-helix protein secondary structures in comparison with the control group.As can be observed,the spectra of burn and control groups were totally separated with PC-1,which explains most of the variability(71.94%).Spectra corresponding to the burn group were characterized by a high intensity of bands at 1624 cm-1 and 1689 cm-1(β-sheet andβ-turn secondary structure of amide I).The constructed PLS-DA classification model for identifying fatal burns demonstrated excellent classification ability with 100%accuracy.Conclusion The preliminary study showed the potential of FTIR in combination with chemometric methods as a supplementary tool for post-mortem diagnosis of fatal burns.
