太白楤木总皂苷对无水乙醇致大鼠胃溃疡的预防作用 被引量:4

摘要 目的观察太白楤木总皂苷(TSAT)对无水乙醇诱导大鼠胃溃疡的预防作用。方法将36只SD大鼠随机分成空白组、模型组、胃康灵组以及低、中、高剂量TSAT组,每组6只。空白组和模型组大鼠均给予生理盐水灌胃,胃康灵组大鼠给予胃康灵胶囊(0. 5 g/kg)灌胃,低、中、高剂量TSAT组大鼠分别给予3、6、12 g/kg TSAT灌胃。末次给药1 h后,除空白组外,其余各组均予10 ml/kg无水乙醇灌胃造模。1 h后,观察各组大鼠胃黏膜大体形态及组织形态学,检测胃液pH值及胃黏膜组织中H^+-K^+-ATP酶活性。结果模型组大鼠胃黏膜呈深红色,肿胀,纵行皱襞可见条索样出血。各剂量TSAT组大鼠胃黏膜出血及损伤程度较模型组减轻。模型组胃液pH值高于空白组(P <0. 05),胃康灵组、中剂量及高剂量TSAT组胃液pH值均低于模型组(均P <0. 05)。模型组胃黏膜组织H^+-K^+-ATP酶活性低于空白组(P <0. 05),胃康灵组及各剂量TSAT组胃黏膜组织H^+-K^+-ATP酶活性均高于模型组(均P <0. 05)。结论 TSAT对无水乙醇致大鼠胃溃疡具有良好的预防作用,其黏膜保护作用可能与提高壁细胞膜H^+-K^+-ATP酶活性、恢复胃液pH值有关。 Objective To observe the protective effect of total saponin of Aralia taibaiensis (TSAT) on gastric ulceRinduced by anhydrous ethanol in rats.Methods Thirty-six SD rats were randomly divided into blank group,model group,Weikangling group,low-dose TSAT group,medium-dose TSAT group,oRhigh-dose TSAT group,with 6 rats in each group.Intragastric administration of normal saline was conducted in the rats in the blank group and the model group.Intragastric administration of Weikangling Capsule(0.5 g/kg) was conducted in the Weikangling group.Intragastric administration of TSAT of 3 g/kg,6 g/kg and 12 g/kg were conducted in the rats in the low-dose TSAT group,medium-dose TSAT group and high-dose TSAT group,respectively.One houRafteRthe last administration,modeling was performed by intragastric administration of anhydrous ethanol in the otheRgroups except the blank group,and 1h later,the gross and histological morphology of gastric mucosa were observed in the rats of each group,the pH of gastric juice and H^+-K^+-ATPase activity of gastric mucosa were detected.Results Gastric mucosa was cardinal and swollen,and stripe-liked bleeding was found in longitudinal plica in the model group.The severity of gastric mucosal bleeding and injuries were mildeRin the TSAT group of all doses than in the model group.The pH value of gastric juice was higheRin the model group than in the blank group( P <0.05),and loweRin the Weikangling group,medium-dose TSAT group and high-dose TSAT group than in the model group(all P <0.05).The H^+-K^+-ATPase activity of gastric mucosa was loweRin the model group compared than in the blank group ( P <0.05),higheRin the Weikangling group and each dose TSAT group than in the model group(all P <0.05).Conclusion TSAT obtains an excellent preventive effect from gastric ulceRinduced by anhydrous ethanol in rats.Its protective effect on mucosa may be related to the increased H^+-K^+-ATPase activity of parietal cells and the normalization of pH environment in gastric juice.
