摘要
目的研究转录因子SOX4对口腔鳞癌(oral squamous cell carcinoma,OSCC)细胞体外生长的影响,以进一步研究SOX4的生物学功能。方法本研究拟以前期研究为基础,利用RNA干扰技术实现SOX4基因敲减,采用细胞计数法、MTT法、细胞划痕和细胞侵袭实验研究转染后的口腔鳞癌细胞UM1体外生长和增殖的变化。结果细胞计数法和MTT法显示:SOX4/siRNA转染后,UM1细胞在48、72h的生长和增殖能力下降(P<0.01);细胞划痕和细胞侵袭实验结果示:SOX4/siRNA转染后,UM1细胞的迁移能力和侵袭能力均下降。结论在RNA干扰实验中发现SOX4在口腔鳞癌UM1细胞中的生长和增殖能力发生明显变化,提示SOX4蛋白可能具有促进口腔鳞癌发生发展的作用。
Objective To study the effect of the transcription factor SOX4 on the cell growth of oral squamous cell carcinoma(OSCC)in vitro so as to further study the biological function of SOX4.Methods The RNA interference technology was used to knock down the expression of SOX4 in the OSCC cells.The methods including cell counting,MTT,cell scratch test and cell invasion test were utilized to examine the growth and proliferation of the transfected OSCC cells UM1 in vitro.Results The results of cell counting and MTT showed that the knockdown of SOX4 significantly inhibited the growth and proliferation of UM1 cells(P<0.01),and those of cell scratch test and cell invasion test showed that their ability of migration and invasion was also decreased.Conclusion It is found in the RNA interference study that SOX4 changes the growth and proliferation of the OSCC cells UM1,which suggests that SOX4 might be actively involved in the progression of OSCC.
