摘要
目的用分子生物学的方法对福尔马林固定液中浸泡的泡球蚴标本进行鉴别诊断。方法从感染多房棘球绦虫的大鼠体内提取泡球蚴病理组织,用福尔马林固定液分别浸泡6个月和12个月,将经浸泡后的组织提取DNA,用多房棘球绦虫的特异性引物EmH15/17进行扩增,与多房棘球绦虫的线粒体12SRNA序列对比,从而进行虫种鉴定。结果在扩增的43例样本中,福尔马林固定液中浸泡6个月和12个月的泡球蚴组织分别有25例和18例,经PCR扩增后比对,发现均与多房棘球绦虫具有相同序列。结论用这种分子生物学方法,可以使保存于福尔马林固定液中鼠来源的泡球蚴标本得到较满意鉴定,预示这种方法对于人体来源的泡球蚴福尔马林固定标本的分子鉴定工作具有可行性。
Objective To differentially diagnose the specimens of alveolar hydatid cyst immersed in formalin fixative by the molecular biological method.Methods The pathological tissues of the alveolar hydatid cyst were extracted from the rats infected with echinococcus multilocularis and soaked in formalin fixative for 6 and 12 months respectively.The DNA extracted from the soaked tissues was amplified with the specific primer EmH15/17 of echinococcus multilocularis and compared with the mitochondrial 12S RNA sequence of echinococcus multilocularis to identify the species.Results Among the 43 amplified samples,there were 25 cases and 18 cases of alveolar hydatid cyst tissues in the formalin fixative for 6 and 12 months respectively.After the PCR amplification,their sequences were identical to the sequence of echinococcus multilocularis.Conclusion This molecular biological method can be used to obtain a satisfactory identification of murine-derived alveolar hydatid cyst stored in formalin fixative,which suggests that this method is feasible for the molecular identification of the formalin-fixed specimens of human-derived alveolar hydatid cyst.
