摘要
为研究牛源cathelicidin类抗菌肽的生物学功能,通过生物信息学方法对牛源cathelicidin类抗菌肽的理化性质、信号肽、跨膜区、亚细胞定位、二级结构和化学修饰等进行预测分析.结果表明:牛源cathelicidin类抗菌肽为阳离子型抗菌肽,等电点11.53~13.20;除了BMAP-27和Dodecapeptide为稳定的多肽外,其他抗菌肽均为不稳定多肽;BMAP-28和Dodecapeptide为疏水性多肽,BMAP-27、BMAP-34、Bac-4、Bac-5、Bac-7和Indolicidin为亲水性多肽;BMAP-27和BMAP-28由α螺旋、β折叠、β转角和无规则卷曲构成,BMAP-34主要由α螺旋和β转角构成,Bac4、Bac7、Indolicidin和Dodecapeptide由无规则卷曲构成,Bac5由β转角和无规则卷曲构成;没有信号肽、跨膜区和糖基化位点,BMAP-27、BMAP-28和BMAP-34各存在1个丝氨酸的磷酸化位点.根据分析结果推测牛源cathelicidin类抗菌肽可作用于细菌的细胞壁或细胞膜,进而导致细菌死亡.
To study the biological functions of bovine cathelicidin antimicrobial peptides(bcAMPs),tools on line were applied to analyze the physical and chemical properties,signal peptide,trans-membrane region,subcellular localization,secondary structure and chemical modification of bcAMPs.The results showed that bcAMPs are cationic antimicrobial peptide,and their isoelectric point are between 11.53 and 13.20.Except that BMAP-27 and Dodecapeptide are stable peptide,other bcAMPs are unstable peptide.BMAP-28 and Dodecapeptide are hydrophobic peptide,while BMAP-27,BMAP-34,Bac-4,Bac-5,Bac-7 and Indolicidin are hydrophilic peptide.BMAP-27 and BMAP-28 have alpha helix,beta sheet,beta turn and random curl,while BMAP-34 is mainly composed of alpha helix and beta turn.Bac4,Bac7,Indolicidin and Dodecapeptide consist of random curl.Bac5 consists of beta sheet and random curl.The bcAMPs have no signal peptide,trans-membrane region and glycosylation sites.BMAP-27,BMAP-28 and BMAP-34 have only one serine phosphorylation site respectively.These findings suggested that bcAMPs could kill bacterial through the interaction with cell wall or cell membrane.
