FGF7亚家族在胚胎期小鼠毛囊形成过程中的差异表达 被引量:1

摘要 为了研究FGF7亚家族(FGF7/FGF10/FGF22)在胚胎小鼠毛囊形成过程中的作用,选取胚胎期(13~17 d)小鼠,采用免疫组织化学和蛋白免疫印迹技术(Western blot)检测小鼠胚胎毛囊形成不同阶段皮肤组织中FGF7/FGF10/FGF22的蛋白表达。结果显示,FGF7/FGF10/FGF22在毛胚细胞、毛钉、球根状毛钉、表皮和真皮细胞中均有表达,但FGF7在14日龄的胚胎皮肤中表达量最高,FGF10在15日龄的胚胎皮肤中表达量最高,FGF22在17日龄的胚胎皮肤中表达量最高。结果提示,FGF7亚家族3个成员在胚胎小鼠毛囊形成过程中均可发挥重要作用,FGF7在毛囊形成的启动阶段即胚胎期第14天发挥主要作用,可能参与指导毛胚细胞的增殖;FGF10在毛囊形成的关键阶段即胚胎期第15天发挥主要作用,可能参与指导毛钉和球根状毛钉的形成;FGF22在毛囊的基本形成阶段即胚胎期第17天发挥主要作用,可能促进毛钉成熟。 The study focuses on the role of FGF7 subfamily(FGF7/FGF10/FGF22)in embryonic mouse hair follicle formation.Immunohistochemistry and Western blot were used to test the protein expression of FGF7/FGF10/FGF22 in the skin tissue during the hair follicle formation stage of mouse embryos on 13-17 day.The results showed that FGF7/FGF10/FGF22 were mainly expressed in hair germs,hair pegs,bulbous hair pegs,epidermal and dermal cells.But the highest protein FGF7 expression was observed on the 14-day-old embryonic skin.Protein expression of FGF10 was the highest on the 15-day-old embryonic skin,whereas the highest protein FGF22 expression was detected on 17-day-old embryonic skin.Experimental results suggest that three members of the FGF7 subfamily play roles in embryonic mouse hair follicle formation.FGF7 plays a major role in the initiation phase of hair follicle formation,that is on the 14th day of embryonic phase,may participate in instructing hair germ cells to proliferate.FGF10 plays a major role in the key stage of hair follicle formation,that is on the 15th day of embryonic phase,may participate in instructing the formation of hair pegs and bulbous hair pegs.FGF22 plays a major role in the basic stage of hair follicle formation,that is on the 17th day of embryonic phase,may promote the maturity of hair pegs.
