摘要
测定不同产地、生长年限、采收期猪苓药材中麦角甾醇、麦角甾-4,6,8(14),22-四烯-3-酮及总多糖3种有效成分的含量;根据不同成分含量的差异,得到最佳猪苓生长年限及最佳猪苓采收期。采用HPLC对麦角甾醇及麦角甾-4,6,8(14),22-四烯-3-酮进行含量测定,以葡萄糖为对照品,采用硫酸-蒽酮法测定猪苓总多糖含量。结果表明,不同产地猪苓药材中麦角甾醇、麦角甾-4,6,8(14),22-四烯-3-酮及总多糖含量存在一定差异,麦角甾醇以河北临城猪苓药材含量最高(1.41 mg/g),麦角甾-4,6,8(14),22-四烯-3-酮以云南泸西猪苓药材中含量最高(18.87 g/g),多糖以吉林猪苓药材中含量最高(4.99%);陕西地区猪苓药材中3种成分含量都普遍高于其他产地,猪苓药材的质量较好;陕西略阳猪苓中麦角甾醇、麦角甾-4,6,8(14),22-四烯-3-酮及总多糖含量随着生长年限的变化而改变;相同生长年限猪苓药材的采收时期选择春季为宜,与传统采收期相符合。此方法具有灵敏、简便、准确的优点,可以为寻找最佳的猪苓产地及猪苓的最佳生长年限及采收期提供科学依据。
The contents of ergosterol, ergosterone-4, 6, 8(14), 22-tetraen-3-one and polysaccharide of Polyporus umbellatus in different growth periods and different harvest periods were determined. The best regions, the best growth period and the best harvest period were obtained according to the difference in the content of different components. Ergosterol and ergosterone-4, 6, 8(14), 22-tetraen-3-one were determined by HPLC, and polysaccharides in P. umbellatus were determined by UV-vis, glucose as reference substance. The results showed that there were some differences in the contents of ergosterol, ergosterone-4, 6, 8(14), 22-tetraen-3-one and polysaccharides in different regions. The highest ergosterol of 1.41 mg/g is from Lincheng, Hebei. The highest ergosterone-4, 6, 8 (14), 22-tetraen-3-one of 18.87 (g/g is from Luxi, Yunnan. The highest polysaccharides of 4.99%is fromJilin. The contents of three components of P. umbellatus in Shaanxi are generally higher than those in other producing areas. In the same area (Lueyang, Shaanxi), ergosterol, ergosterone-4, 6, 8(14), 22-tetraen-3-one and polysaccharides in P. umbellatus changed with the growth of the growth period. For the same growth period, the harvest time of P. umbellatus is appropriate in spring. This method is simple, sensitive and accurate. It provides scientific basis for finding the best place of regions, the best growth period and the best harvest period.
