摘要
目的探讨分析女贞子乙醇提取物对人毛囊无色素黑素细胞c-kit表达的影响。方法取体外培养的人毛囊无色素黑素细胞,以不同浓度的女贞子乙醇提取物进行干预,采用显微镜观察和细胞数统计的方法测定女贞子乙醇提取物对无色素黑素细胞增殖的影响,选取最佳浓度。RT-PCR测定最佳浓度下c-kit mRNA的相对表达量,并进行统计分析。结果女贞子提取物可促进毛囊无色素黑素细胞的增殖,最佳浓度为0.22mg/mL,并可对c-kitm RNA表达有显著的促进作用。结论女贞子乙醇提取物可促进体外培养的人毛囊无色素黑素细胞的增殖和c-kit的表达。
Objective To investigate the effect of ligustrum lucidum ethanol extract on c-kit expression in human hair follicle amelanotic melanocyte.Method Human hair follicle amelanotic melanocytes were cultured in vitro,and melanocyte culture was carried out with different concentration of Ligustrum lucidum extract.The effects of Ligustrum lucidum extract on the proliferation of amelanotic melanocytes were studied by microscopy and cell counting.The relative expression of C-kit mRNA at the optimal concentration was determined by RT-PCR,and the statistical analysis was carried out.Result Ligustrum lucidum extract can effectively promote the proliferation of amelanotic melanocytes.It can significantly promote the expression of C-kit mRNA at the optimum concentration was 0.22 mg/mL.Conclusion Ligustrum lucidum ethanol extract can effectively promote cell proliferation and c-kit expression for human hair follicle amelanotic melanocytes cultured in vitro.
