摘要
目的探讨P21蛋白激活激酶1(P-21 activated kinase 1,PAK1)在非小细胞肺癌的表达,以及下调PAK1基因对细胞增殖和侵袭的影响。方法选取非小细胞肺癌患者97例,实时荧光定量PCR检测非小细胞肺癌和癌旁组织中PAK1基因表达,培养非小细胞肺癌A549细胞株,分为PAK1干扰组、阴性对照组和空白组,实时荧光定量PCR检测细胞中PAK1基因表达,MTT法检测细胞增殖活力,Transwell法检测细胞侵袭能力,Western blot法检测细胞中PAK1、β-catenin、磷酸化β-catenin和MMP-9蛋白表达。结果非小细胞肺癌组织中PAK1 mRNA相对表达量高于癌旁组织,差异有统计学意义(t=52. 063,P=0. 000);非小细胞肺癌组织中PAK1 mRNA相对表达量与TNM分期、分化程度和淋巴结转移有关(P <0. 05); PAK1干扰组细胞中PAK1 mRNA相对表达量低于阴性对照组和空白组(P <0. 05); PAK1干扰组细胞48h、72h和96h吸光度A值低于阴性对照组和空白组(P <0. 05); PAK1干扰组侵袭细胞数低于阴性对照组和空白组(P <0. 05); PAK1干扰组PAK1、磷酸化β-catenin和MMP-9蛋白相对表达量低于阴性对照组和空白组,而β-catenin蛋白相对表达量高于阴性对照组和空白组(P <0. 05)。结论 PAK1在非小细胞肺癌组织中呈高表达,下调PAK1基因表达可抑制非小细胞肺癌细胞增殖和侵袭,其机制可能与β-catenin信号通路有关。
Objective To investigate the expression of PAK1 in non-small cell lung cancer tissue,and to explore the effect of down-regulation of PAK1 gene on cell proliferation and invasion.Methods 97 patients with non-small cell lung cancer were selected.Real-time fluorescence quantitative PCR was used to detect the expression of PAK1 gene in non-small cell lung cancer and adjacent tissues.The non-small cell lung cancer A549 cell line was cultured and was divided into the PAK1 interference group,the negative control group and the blank group.Real-time fluorescence quantitative PCR was used to detect the expression of PAK1 gene in cells.MTT assay was used to detect cell proliferation.Transwell assay was used to detect cell invasion.Western blot analysis was used to detect the expressions of PAK1,β-catenin,phospho-β-catenin and MMP-9 proteins in cells.Results The relative expression level of PAK1 mRNA in non-small cell lung cancer tissue was higher than that in adjacent tissue(t=52.063,P=0.000).The relative expression level of PAK1 mRNA in non-small cell lung cancer tissue was related with TNM stage,differentiation and lymph node metastasis(P<0.05).The relative expression level of PAK1 mRNA in the PAK1 interference group was lower than that in the negative control group and the blank group(P<0.05).The absorbance A value at 48h,72h and 96h in the PAK1 interference group was lower than that in the negative control group and the blank group(P<0.05).The number of invasive cells in the PAK1 interference group was lower than that in the negative control group and the blank group(P<0.05).The relative expression levels of PAK1,phosphorylatedβ-catenin,and MMP-9 proteins in the PAK1 interference group were lower than those in the negative control group and the blank group,while the relative expression level ofβ-catenin protein was higher in the PAK1 interference group than that in the negative control group and the blank group(P<0.05).Conclusion PAK1 is highly expressed in non-small cell lung cancer tissue.Down-regulation of PAK1 g
