摘要
目的观察抗结核药链霉素-硫酸钙颗粒在兔子体内的缓释性能,评估安全性。方法选取新西兰大白兔48只,将其随机分为两组,每组24只。实验组在胫骨近端骨缺损置入链霉素-硫酸钙颗粒;对照组植入等量的链霉素。采用高效液相色谱(HPLC)仪测定链霉素浓度。术后不同时间点抽血待检;孔周围取组织待检。术后2、6周行肝肾抽血检查及肝、肾脏组织病理切片检查。植入部位行X线检查。结果实验组中链霉素在周围组织维持9周最低杀菌浓度(MBC),植入10周尚处于最低抑菌浓度(MIC)。静脉血液中可维持8周MBC,植入9周尚处于MIC。对照组链霉素在周围组织维持2周MBC,植入3周处于MIC;在血液维持1周MBC,植入2周处于MIC。实验组血液和组织的药物浓度高于对照组,差异均有统计学意义(P<0.05)。肝脏、肾脏功能生化检查、病理切片示安全性良好,两组肝肾功能组间比较,差异均无统计学意义(P>0.05)。X线及病理学检查示实验组具有良好的成骨性。结论链霉素-硫酸钙颗粒在兔子体内缓慢释放,达到缓释效果,对肝肾组织无损伤。
Objective To observe the sustained release property of the anti-tuberculosis drug-Streptomycin-Calcium Sulfate Particles in rabbits and evaluate the safety.Methods All of 48 white rabbits of New Zealand were selected and they were randomly divided into the two groups,and each group had 24 rabbits.In the experimental group,Streptomycin-Calcium Sulfate Particles were placed in the proximal tibial bone defect while the equivalent Streptomycin was implanted in the control group.The concentration of Streptomycin was determined by high performance liquid chromatography instrument.Blood samples were taken at different time points after operation and tissues around the foramen were taken for examination.Blood samples of liver and kidney and histopathological sections of liver and kidney were examined at 2 and 6 weeks after operation.X-ray examination was performed at the implant site.Results In the experimental group,the lowest bactericidal concentration(MBC)was maintained in the surrounding tissue for 9 weeks,and the minimum inhibitory concentration(MIC)was still 10 weeks after implantation.The MBC was maintained in venous blood for 8 weeks,and MIC was still 9 weeks after implantation.In the control group,the MBC of Streptomycin in the surrounding tissues was maintained for 2 weeks,and the MIC was still after 3 weeks of implantation and the MBC of Streptomycin in the blood was maintained for 1 week and MIC was still after 2 weeks implantation.The drug concentration of blood and tissue in the experimental group was higher than that in the control group,and the differences were statistically significant(P<0.05).Liver and kidney function biochemical examination and pathological biopsy showed good safety and no toxic reaction.There was no significant difference between the two groups(P>0.05).X-ray and pathological examination showed that the experimental group had good osteogenesis.Conclusion Streptomycin-Calcium Sulfate Particles are released slowly in rabbits to achieve a sustained release effect without damage to liver a
