摘要
目的探讨针刺夹脊穴对大鼠胃肠动力障碍的作用及机制。方法将45只SD大鼠随机分为空白对照组、模型组、针刺组、针刺+N-甲基-D-天冬氨酸受体(NMDAR)拮抗剂组、针刺+γ-氨基丁酸(GABA)受体拮抗剂组,每组9只。除空白对照组外其他各组皮下注射硫酸阿托品注射液建立胃肠动力障碍模型。针刺+NMDAR拮抗剂组给予迷走神经背核(DMV)微量注射犬尿烯酸,针刺+GABAR拮抗剂组给予荷苞牡丹碱、法克罗芬,空白组、模型组、针刺组注射无菌水。微量注射10 min后,针刺组、针刺+NMDAR组、针刺+GABAR拮抗剂组给予夹脊穴针刺,并接电针刺激30 min。针刺结束10 min,处死大鼠,取胃及小肠组织,计算胃排空率、小肠推进率。结果与空白对照组比较,模型组胃排空率、小肠推进率低(P均<0. 01),针刺组胃排空率、小肠推进率较模型组高(P均<0. 05)。针刺+GABAR拮抗剂组与针刺组比较差异无统计学意义(P均> 0. 05)。针刺+NMDAR组胃排空率、小肠推进率较针刺组低(P均<0. 05)。结论针刺夹脊穴可促进大鼠胃肠运动,其作用机制可能与DMV中的NMDAR有关。
Objective To observe the effect and mechanism of Jiaji acupoint on gastrointestinal motility disorder in rats. Methods Forty-five SD rats were randomly divided into the control,model,acupuncture (ACU),ACU + NMDAR anatagonist,ACU +γ-aminobutyric acid (GABAR)antagonist groups,with 9 rats in each group. The rats were subcutane-ously injected with atropine to make the models of gastrointestinal dyskinesia except the control group. The rats in the ACU, ACU + NMDAR antagonist,ACU + GABAR antagonist groups were given Jiaji acupuncture. The rats in the ACU + NMDAR antagonist group were microinjected with the NMDA receptor (NMDAR)antagonist kynurenic acid,and the rats in the ACU + GABAR antagonist group were microinjected with the GABA receptor(GABAR)antagonist dicentrine and phaclofen in the dorsal motor nucleus of vagus (DMV),respectively after acupuncture. The rats in the normal control group were giv-en distillated water. After 10 minutes of microinjection,rats in the ACU,ACU + NMDAR anatagonist,ACU + GABAR anatagonist groups were given Jiaji acupoint electro-acupuncture for 30 minutes. The rates of gastric emptying and small bowel peristalsis were calculated. Results Compared with the control group,the gastric emptying rate and small bowel peristalsis rate in the model group were lower (both P <0.01),while the gastric emptying rate and small intestinal peristal-sis rate in the ACU group were higher than those of the model group (both P <0.05). There was no significant difference in the gastric emptying rate and small intestinal peristalsis rate between the ACU + GABAR antagonist group and the ACU group (both P >0.05). The gastric emptying rate and small intestinal peristalsis rate in the ACU + NMDAR antagonist group were lower than those in the ACU group (P < 0.05). Conclusion Jiaji acupoint could improve the gastrointestinal motility disorder of rats,which may be related to the NMDA receptors of DMV.
