摘要
马尾松(Pinus massoniana)是我国南方重要的用材树种。为丰富马尾松基因组信息,弄清松属植物内部亲缘关系,本研究通过高通量测序技术对马尾松叶绿体基因组进行测序和组装,并对其进行特征分析和聚类关系研究。生物信息学分析结果表明,马尾松叶绿体基因组序列全长119 743 bp,GC含量为38.54%;共注释得到126个基因,包括84个蛋白编码基因,38个tRNA基因和4个r RNA基因,其结构无典型的反向重复序列;共检测到15个散在重复序列和26个串联重复序列;蛋白编码基因对应的密码子偏好使用A/T碱基,并且编码亮氨酸的密码子使用频率最高,编码半胱氨酸的密码子数最少。系统发育研究表明,马尾松与拉雅松(Pinus crassicorticea)亲缘关系最近,与湿地松(Pinus elliottii)和两个南亚松(Pinus latteri)亲缘关系较远。研究结果建立了适于松属植物完整叶绿体基因组组装及其特征分析的方法,对叶绿体基因组工程研究及松属种间的分子标记开发具有一定的参考价值,为松属植物的进化和系统发育提供方法指导。
The high-throughput sequencing technology was used to sequence and assemble the chloroplast genome of Pinus massoniana,and its characteristics and cluster relationships were studied.Results showed that the total sequence of chloroplast genome of P.massoniana was 119743 bp,with GC content of 38.54%.126 genes were annotated,including 84 protein-coding genes,38 tRNA genes,and 4 rRNA genes.The structure was not typical reverse repetitive sequence.Furthermore,the cp genome of Pinus massoniana contained 15 dispersed repeat sequences and 26 tandem repeat sequences.The codon usage of protein-coding genes was biased to use A/T bases,and among these codons,leucine and cysteine were the most and least used amino acids,respectively.The cluster analysis showed that P.massoniana had a closer genetic relationship with Pinus crassicorticea,and it had a long association with Pinus elliottii and two species of Pinus latteri.The results established a method for genomic assembly and characterization of intact chloroplasts of Pinus,which has certain reference value for the study of chloroplast genome engineering and the development of molecular markers between Pinus,providing a methodological guidance for evolution and phylogenetic analysis of Pinus in the future.
