不同造模方法对骨关节炎肾阳虚证动物模型相关指标的影响 被引量:8

Influence of the different modeling methods on the related indexes in the animal models with osteoarthritis of kidney yang deficiency

摘要 目的检测不同造模方法下,骨关节炎(Osteoarthritis,OA)肾阳虚证模型大鼠相关指标的差异,以利于OA肾阳虚证造模方法的选择。方法将45只SD大鼠按造模方法随机分为正常组(不干预)、模型组(hulth法干预)、D半乳糖组(hulth法及D半乳糖干预)、腺嘌呤组(hulth法及腺嘌呤干预)、去势组(hulth法及去势干预)。造模成功后检测大鼠证候学评分、脏器指数、皮质酮(Corticosterone,CORT)、雌二醇(Estradiol,E2)、睾酮(Testosterone,T)含量,对大鼠关节进行大体观察、HE染色和Mankin's评分,记录大鼠死亡情况。结果腺嘌呤组及去势组在证候学评分、脏器指数(去势组除外)及肾阳虚病理学指标(CORT、E2、T)方面,均较正常组升高,差异有统计学意义(P <0. 05),二者均满足肾阳虚证候标准; D半乳糖组、腺嘌呤组、去势组3组在关节大体形态、HE染色方面,均表现出OA特征,Mankin's评分较正常组得分高,差异有统计学意义(P <0. 05),三者均符合OA的模型特点。造模成功后,腺嘌呤组死亡4只,其余组无死亡。结论腺嘌呤法或去势法联合hulth法造模,成模后模型既符合肾阳虚证的表现也符合OA的表现,然腺嘌呤法联合hulth法造模动物死亡率高,故去势法联合hulth法更为适合OA肾阳虚证的模型建立。 Objective To determine the differences in the related indexes in the rat models with osteoarthritis(OA) of kidney yang deficiency with different modeling methods so as to optimize the modeling method for OA of kidney yang deficiency syndrome. Methods According to the modeling methods,45 SD rats were divided into a normal group(without intervention),a model group(with Hulth intervention),a D-glactose group(with Hulth method and D-glactose),an adenine group(with Hulth method and adenine) and a castration group(with with Hulth method and castration).After successful modeling,the symptom score,visceral indexes and the levels of corticosteron(CORT),estradil(E2) and testosterone(T) were determined. The joint gross observation,HE staining and Mankin’ s core were performed in the rats.The mortality was recorded in the rats. Results In the adenine group and the castration group,the symptom score,the visceral indexes(except the castration group) and the pathological indexes(CORT,E2 and T) of kidney yang deficiency were all increased as compared with the normal group,indicating the significant differences(P < 0. 05),which was in compliance with the criteria of kidney yang deficiency syndrome. In the D-glactose group,the adenine group and the castration group,the joint gross morphology and HE staining all showed the OA characteristics and Mankin’s score was higher than the normal group,indicating the significant differences(P < 0. 05),which was in compliance with the OA modeling characteristics. After successful modeling,in the adenine group,4 rats were dead and no rat was dead in the rest groups. Conclusion With the intervention with adenine or the castration combined with Hulth method,the modeled rats are in compliance with the manifestations of kidney yang deficiency and OA. The mortality is high with the intervention of adenine combined with Hulth method. Hence,the castration combined with Hulth method is more suitable for the modeling of OA of kidney yang deficiency.
