摘要
目的探讨钙激活蛋白43(Cap43)在胶质瘤组织中表达,以及下调Cap43表达对人胶质瘤U251细胞增殖、迁移和侵袭的影响。方法选取2014年2月至2017年3月在我院经手术切除的初发患者91例,同期,选取因颅脑外伤切除的正常脑组织40例作为对照组,免疫组化法检测胶质瘤组织和对照组中Cap43蛋白表达,培养U251细胞并分为Cap43干扰组、对照序列组和空白组,Western blot检测细胞中Cap43蛋白表达,CCK8法检测细胞增殖,Transwell法检测细胞迁移和侵袭能力,构建裸鼠移植瘤模型,4周后,比较各组瘤体重量和体积。结果胶质瘤组织Cap43蛋白的IOD值低于对照组,差异有统计学意义(t=64. 055,P=0. 000);胶质瘤组织中Cap43蛋白的IOD值在不同WHO分级间差异有统计学意义(P <0. 05); Cap43干扰组细胞中Cap43 mRNA和蛋白相对表达量低于对照序列组和空白组(P <0. 05); Cap43干预组细胞在1~4d时吸光度A值均高于对照序列组和空白组(P <0. 05); Cap43干扰组迁移细胞数和侵袭细胞数均高于对照序列组和空白组(P <0. 05); Cap43干扰组裸鼠移植瘤重量和体积均高于对照序列组和空白组(P <0. 05)。结论 Cap43蛋白在胶质瘤组织中呈低表达,且与肿瘤恶性程度负相关,下调Cap43表达可促进细胞增殖、加速细胞迁移和侵袭,并可促进裸鼠移植瘤生长。
Objective To investigate the expression of calpain 43 in glioma tissue,and the effect of down-regulating the expression of Cap43 on human glioma U251 cells proliferation,migration and invasion.Methods Total 91 newly diagnosed patients who underwent surgical resection in our hospital from February 2014 to March 2017 were selected.In the same period,40 cases of normal brain tissues resected from craniocerebral trauma were selected as the control group.The expressions of Cap43 proteins in glioma tissues and control group were detected by immunohistochemistry.U251 cells were cultured and were divided into Cap43 interference group,control sequence group and blank group.Western blot was used to detect the expression of Cap43 in cells.CCK8 method was used to detect cell proliferation.Transwell assay was used to detect cell migration and cell invasion.Xenograft glioma tumor in nude mice model was constructed.After 4 weeks,the weight and volume of Xenograft glioma tumor in each group were compared.Results The IOD value of Cap43 protein in the glioma tissues was lower than that in the control group,the difference was statistically significant(t=64.055,P=0.000).The differences of the IOD values of Cap43 proteins in glimo tissues between different WHO grades were statistically different(P<0.05).The relative expression levels of Cap43 mRNA and protein in Cap43 interference group was lower than that in the control sequence group and blank group(P<0.05).The absorbance A values at 1~4 d in the Cap43 interference group were higher than those in the control sequence group and blank group(P<0.05).The number of migrating cells and the number of invasive cells in the Cap43 interference group were higher than those in the control sequence group and blank group(P<0.05).The weight and volume of nude mice Xenografts in the Cap43 interference group were higher than those in the control sequence group and blank group(P<0.05).Conclusion Cap43 protein was lowly expressed in the glioma tissue,and was negatively correlated with the degree of t
