红缘拟层孔菌多糖提取工艺的响应面优化 被引量:6

摘要 【目的】优化红缘拟层孔菌多糖的超声提取工艺,为其工业化生产和综合利用提供依据。【方法】以红缘拟层孔菌为材料,考察超声功率、提取温度、液(mL)料(g)比和提取时间对红缘拟层孔菌多糖得率的影响;在此基础上,通过Box-Behnken响应面法,设计4因素3水平试验,建立多糖提取回归方程,确定其最佳提取工艺。【结果】超声功率、提取温度、液(mL)料(g)比和提取时间4个因素对红缘拟层孔菌多糖提取的影响大小依次为:提取温度>超声功率>提取时间>液料比,在单因素试验及响应面优化基础上,确定红缘拟层孔菌多糖的最佳提取工艺为:超声功率60W,提取温度63℃,液料比21∶1,提取时间32min;在此条件下,多糖得率为12.87%,纯度为69.42%。【结论】得到了红缘拟层孔菌多糖超声提取的优化工艺,该工艺方便可行。 【Objective】Ultrasonic extraction of polysaccharides from Fomitopsis pinicola was optimized to provide basis for its development and application.【Method】The effects of ultrasonic power extraction,temperature,liquid(mL)-solid(g)ratio and extraction time on yield of F.pinicola polysaccharide were investigated.Then,experiments were designed based on Box-Behnken response surface with 4 factors and 3 levels,and the regression equation of polysaccharide extraction was established to determine the optimum extraction conditions.【Result】The effects of the 4 factors on the extraction of polysaccharides from red marginal bacteria were in the order of ultrasonic temperature>power extraction>extraction time>liquid(mL)-solid(g)ratio.On the basis of single factor test and response surface optimization,the optimum extraction conditions for polysaccharide extraction from red border were ultrasonic power 60 W,extraction temperature 63℃,solid to liquid ratio 21∶1,and extraction time 32 min.The obtained yield of polysaccharides was 12.87%and the purity was 69.42%.【Conclusion】The optimal ultrasonic extraction technology for polysaccharides from F.pinicola was convenient and feasible.
