O-糖苷键连接的N-乙酰葡糖胺(O-GlcNAc)修饰是在许多亚细胞结构发现的蛋白质的丝氨酸和苏氨酸残基的翻译后修饰方式,O-GlcNAc修饰逐渐成为许多心血管病理生理过程的关键介质。早期研究表明提高蛋白质O-GlcNAc修饰水平会导致糖尿病相关的心血管并发症,但随后发现,短期内急剧增加O-GlcNAc修饰水平却对缺血/再灌注损伤具有保护作用。蛋白质的O-GlcNAc修饰会影响细胞的许多功能,包括转录、蛋白质周转、钙处理以及生物能量学方面。蛋白质O-GlcNAc修饰对于维持正常细胞功能和体内平衡是必需的。由于应激或疾病导致的O-GlcNAc水平变化对心脏的影响是非常复杂的。
O-linkedβ-N-acetylglucosamine(O-GlcNAc)is a post-translational modification of the serine and threonine residues of proteins found in many subcellular locations.O-GlcNAc is becoming a key mediator of a number of cardiovascular pathophysiological processes.Early studies have shown that increased protein O-GlcNAcylation leads to cardiovascular complications associated with diabetes,but subsequent studies have shown that acute increase in O-GlcNAc levels has a protective effect on ischemia-reperfusion injury.It is now increasingly understood that the O-GlcNAc modification of proteins affects many cellular functions,including transcription,protein turnover,calcium processing,and bioenergetics.The role of protein O-GlcNAc modification in the cardiovascular system has been found to be essential for maintaining normal cellular function and homeostasis.The impact of O-GlcNAc cycling changes on the heart due to stress or disease is very complex.
