摘要
目的探求优化的原代培养乳鼠心肌细胞方法。方法 2017年6月1日—2017年6月2日选取泉州市医学高等专科学校提供6只出生24 h内Wistar乳鼠,采用2种不同消化心肌组织的方法,对照组予0.125%胰酶+0.05%I型胶原酶+0.1%Ⅱ型胶原酶逐次消化心肌组织;试验组予0.05%I型胶原酶+0.1%Ⅱ型胶原酶消化数次后再单予0.125%胰酶快速消化心肌组织;比较两组取得的心肌细胞数量以及存活率,免疫荧光测肌钙蛋白鉴定纯度。结果 2种方法均得到心肌细胞数量多[对照组(103.3±4.4),试验组(105.8±5.0)],差异无统计学意义(t=-0.911,P=0.384);细胞即刻存活率试验组明显高于对照组,试验组达91.87%,对照组即刻存活率仅68.07%,差异有统计学意义(χ~2=81.0,P=0.025)。试验组存活率明显高于对照组。结论再改良方法提取心肌细胞,可获得存活率高、活力好的心肌细胞。
Objective To explore the optimal culture method of myocardial cell culture in milk rats.Methods6Wistar neonatal rats born within24h in the Quanzhou Medical College from June1,2017to June2,2017were selected and divided into two groups according to the different digesting myocardial tissue methods,The control group was given0.125%pancreatic enzyme+0.05%I type collagenase+0.1%collagen enzyme to digest myocardial tissue successively,the trial group were given0.05%type I collagenase type+0.1%Ⅱcollagenase digestion for several timesc,then using0.125%pancreatic enzymes quickly digested myocardial tissue.The number and survival rate of myocardial cells obtained from the two groups were compared,and the purity was determined by immunofluorescence.Results Both methods obtained a large number of cardiac cells,(103.3±4.4)in the control group,(105.8±5.0)in the experimental group),and the difference was not statistically significant(t=0.911,P=0.384).The immediate cell survival rate of the experimental group was higher than that of the control group,which reached(91.87%vs68.07%)in the control group and in the test group.The difference was statistically significant(χ^2=81.0,P=0.025),and the survival rate in the test group was obviously higher than that in the control group.Conclusion The remodification method in extracting the cardiac muscle cells can obtain a high cardiac muscle cells with high survival rate and good vitality.
