大豆卵磷脂替代卵黄的稀释液低温保存绵羊精液效果 被引量:5

摘要 为研究大豆卵磷脂稀释液对绵羊精液低温保存效果,用5个浓度的大豆卵磷脂(A1,1. 25%; A2,2. 5%; A3,5%;A4,7. 5%; A5,10%)替代TRIS专利稀释液(C组)中的卵黄进行绵羊精液低温保存试验,在保存第0 d、1 d、3 d、6 d、9 d、12 d对精子活率和顶体完整率进行检测,大豆卵磷脂稀释液效果最优组和C组的保存第9 d的精液进行人工授精试验。结果表明:A1组精液保存效果自保存第6 d起显著高于其他添加组(P <0. 05),且在保存第9 d精子活率为0. 53±0. 06,符合鲜精输精国家标准; A1组与C组在各时间点精子活率、顶体完整率差异均不显著(P> 0. 05),且A1组与C组精子在第9 d活率都高于0. 5,顶体完整率高于60%; A1组与C组的受胎率64. 3%(72/112)、65. 6%(80/122)差异不显著(P> 0. 05)。证明添加1. 25%浓度大豆卵磷脂的稀释液能够进行绵羊精液低温长时间有效保存。 In this experiment,it was aimed at the effect of containing soybean lecithin dilution without yolk for low temperature preservation of sheep semen.Sheep semen low temperature preservation test was carried out with5concentrations of soybean lecithin(group A1,1.25%;group A2,2.5%;group A3,5%;group A4,7.5%;group A5,10%)instead of yolk in TRIS patent dilution(group C).Sperm motility rate and acrosome integrity rate were detected on the0th,1st,3rd,6th,9th,12th day after preservation.The 9th day Semen of the best effective group for soybean lecithin and Group C were carried out Artificial insemination.The results showed that group A1was much higher than other groups since the 6th day for effect of semen preservation(P<0.05),and the sperm motility rate on the 9th day was0.53±0.06,which is in line with the national standard of fresh sperm insemination.There was no significant difference in Sperm motility rate and acrosome integrity rate between group A1and group C at all time points(P>0.05).The Sperm motility rate of group A1and group C was greater than0.5on the9th day and the acrosome integrity rate was higher than60%.The conception rate of group A1and group C was 64.3%(72/112),and 65.6%(80/122)had no significant difference(P>0.05).It was proved that the diluent containing 1.25% soybean lecithin could effectively preserve Sheep Semen at low temperature for a long time.
