摘要
以获得降酸酵母菌Yds-10最佳培养基为目的,以OD600值作为评价指标,对常用五种培养基进行比较,筛选出基础培养基;采用单因素和正交试验对筛选出的基础培养基成分葡萄糖、酵母浸粉、蛋白胨、胰蛋白胨进行优化,得出培养基的最佳组合为:葡萄糖30g/L、酵母浸粉2.0 g/L、蛋白胨2. 0g/L、胰蛋白胨0 g/L。此培养基培养降酸酵母菌Yds-10,其培养液的OD600值比YPD培养基高大4. 1%。
The test is to gain the best medium of acid reducing yeasts(Yds-10).Using OD600value as the evaluation index,we elected best medium with comparing five typical used medium.By single factor and orthogonal test,the compositions would be optimized involved glucose,yeast extract powder,peptone and tryptone.It could be concluded that the best optimum combination of culture medium:glucose30g/L,yeast extract powder2.0g/L,peptone2.0g/L,tryptone0g/L.This medium is cultivated Yds-10yeasts,of which OD600values was about4.1%higher than the YPD medium.
