摘要
目的探讨载脂蛋白(apolipoprotein,Apo)E基因在高血压患者后代中的表达情况及其对血脂浓度的影响。方法选取2015年6月至2016年6月来重钢总医院进行健康体检的168名正常者,其父母为高血压患者的为高血压子代人群组(80名),其余为对照组(88名)。检测ApoE基因在高血压患者后代中的表达情况,并探讨ApoE基因对血脂浓度的影响。结果高血压子代组中,E4/4基因型的比例高达75.00%,E2/2的比例最低为1.25%,对照组中E3/3基因型的比例最高为36.36%。高血压子代组的等位基因E4比例高于对照组,差异有统计学意义(χ~2=23.81,P<0.05)。高血压子代人群等位基因E4会对三酰甘油(triacylglycerol,TG),高密度脂蛋白胆固醇(high density lipoprotein-cholesterol,HDL-C)、ApoA等血脂指标产生影响。E4/4基因型对血脂的影响最大,TG浓度升高、HDL-C浓度降低,与其他基因型的TG、HDL-C浓度变化比较,差异有统计学意义(均P<0.05)。高血压子代组的3种等位基因型均可增高TG浓度;E3等位基因可增高总胆固醇(total cholesterol,TC)、低密度脂蛋白胆固醇(low density lipoprotein-cholesterol,LDL-C)浓度、降低HDL-C和ApoA浓度;E4等位基因可升高ApoB浓度、降低HDL-C浓度,与对照组比较,差异有统计学意义(均P<0.05)。结论高血压子代人群中ApoE基因中异常率较高的为E4,拥有该基因的患者具有较高的血脂异常率,且患高血压的可能性较大。
Objectives To investigate the expression of apolipoprotein(Apo)E gene in offspring of patients with hypertension and its effects on serum lipid concentration.Methods A total of168normal subjects were enrolled in Chongqing Iron and Steel General Hospital from June2015to June2016.The subjects whose parents were with hypertension were divided into offspring group(80cases)and the rest were healthy group(88cases).The expression of ApoE gene in progeny of patients with hypertension was detected and the effect of ApoE gene on serum concentration of lipid was investigated.Results In the offspring of patients with hypertension,the ratio of E4/4genotype was the highest,E2/2ratio was the lowest,and the ratio of E3/3genotype in healthy group was the highest.The E4ratio of offspring group was higher than that of healthy group(χ2=23.81,P<0.05).E4allele of offspring of patients with hypertension had influence on concentrations of triacylglycerol(TG),high density lipoprotein-cholesterol(HDL-C)and ApoA.The E4/4genotype had the greatest effect on blood lipid concentration,HDL-C significantly decreased,TG significantly increased,and the differences were statistically significant when compared with those of the other genotype(P<0.05).The three alleles of the offspring group could increase concentration of TG,E3allele could increase concentrations of total cholesterol(TC),low density lipoprotein-cholesterol(LDL-C),and decrease concentrations of HDL-C and ApoA.E4allele can increase concentration of ApoB and reduce concentration of HDL-C,and the changes of the above indexes were statistically differ?ent from those of healthy group(all P<0.05).Conclusions E4gene has higher abnorma rate in offspring of parents with hypertension,who are more likely to have abnormal blood concentrations of lipids and hypertension.
