摘要
目的探讨加味温胆方对心肌缺血(痰浊血瘀证)大鼠细胞凋亡及B淋巴细胞瘤(Bcl-2)、Bcl-2同源二聚体X蛋白(Bax)、Survivin、Caspase-3表达的影响。方法清洁级SD大鼠雄性100只,根据体质量随机分成5组:空白组、模型组、加味温胆方低剂量组(5.0 mg/kg)、加味温胆方中剂量组(10.0 mg/kg)、加味温胆方高剂量组(20.0 mg/kg)。模型组、加味温胆方各剂量组喂以高脂饲料,同时分别以0.9%氯化钠注射液及各剂量加味温胆方灌胃,连续灌胃7周,在第49天施冠状动脉结扎术;空白组喂以普通饲料,以0.9%氯化钠注射液灌胃,实验结束后,测定凋亡光密度及面积、Bcl-2、Bax、Survivin、Caspase-3蛋白及m RNA水平。结果与空白组比较,模型组及加味温胆方组各剂量Bcl-2、Survivin mRNA、蛋白表达水平降低,Bax、Caspase-3mRNA、蛋白表达水平、心肌细胞凋亡光密度及面积升高(P <0.05);与模型组比较,加味温胆方组各剂量组Bcl-2、Survivin mRNA、蛋白表达水平降低升高,Bax、Caspase-3 m RNA、蛋白表达水平、心肌细胞凋亡光密度及面积降低,且随着加味温胆方组给药剂量的增加,Bcl-2、Survivin mRNA、蛋白表达水平逐渐升高,Bax、Caspase-3 mRNA、蛋白表达水平、心肌细胞凋亡光密度及面积逐渐降低,剂量-效应关系明显(P <0.05)。结论加味温胆方能减轻冠心病大鼠的病理损伤,抑制心肌缺血(痰浊血瘀证)大鼠细胞凋亡,其机制与加味温胆方能促进Bcl-2、Survivin mRNA、蛋白的表达,抑制Bax、Caspase-3 m RNA、蛋白的表达有关。
Objective:To investigate effect of Jiawei Wendan Decoction on cell apoptosis and expression levels of Bcl-2,Bax,Survivin and Caspase-3in myocardial ischemia rats with blood stasis syndrome.Methods:100male SD rats were randomly divided into5groups according to body weight:the blank group,the model group,Jiawei Wendan Decoction low-dose group(5.0mg/kg),middle-dose group(10.0mg/kg)and high-dose group(20.0mg/kg).The model group and Jiawei Wendan Decoction groups were fed with high-fat diet,and the rats were given intragastric administration of normal saline and various doses of Jiawei Wendan Decoction,respectively for7weeks.Coronary artery ligation was performed on the49th day.The blank group was fed with normal diet.After the experiment,the optical density and area of apoptosis and protein expression and mRNA levels of Bcl-2,Bax,Survivin and Caspase-3were measured.Results:Compared with the blank group,mRNA and the expression levels of Bcl-2and Survivin in the model group and Jiawei Wendan Decoction groups were decreased,and that of Bax and Caspase-3and the apoptosis optical density and area of cardiomyocytes were increased(P<0.05).Compared with the model group,mRNA and the expression level of Bcl-2and Survivin in Jiawei Wendan Decoction groups decreased,and that of Bax and Caspase-3,the optical density of cardiomyocyte apoptosis and area were increased.In all indexes,dose-effect relationship was significant(P<0.05).Conclusion:Jiawei Wendan Decoction can reduce the pathological damage of coronary heart disease rats,inhibit the apoptosis in myocardial ischemia rats with blood stasis syndrome,which is related with the upregulation of mRNA and protein expression levels of Bcl-2and Survivin as well as the downregulation of mRNA and protein expression levels of Bax and Caspase-3.
