摘要
目的建立天麻Gastrodia elata BL.水提物HPLC指纹图谱,对其真伪进行鉴别。方法天麻水提取液的分析采用Phenomenex Kinetex C18100柱(250 mm×4. 6 mm,5μm);柱温25℃;流动相乙腈-水(含0. 1%甲酸),梯度洗脱;体积流量1. 0 mL/min;检测波长270 nm,对结果进行相似度和主成分分析。结果 30批天麻水提物指纹图谱中有16个共有峰,相似度均大于0. 800,混淆品相似度较低,分别为0. 029、0. 016、0. 001、0. 175、0. 041、0. 150、0. 004,可以鉴别天麻真伪;主成分分析筛选出11个差异性指标成分,表明天麻指纹成分具有一致性。结论该方法稳定可靠,可用于天麻的质量控制。
AIM To establish the HPLC fingerprints of aqueous extract from Gastrodia elata BL. and to study their identification. METHODS The analysis of aqueous extract from G. elata was developed on a 25 ℃ thermostatic Phenomenex Kinetex C18100 column(250 mm × 4. 6 mm,5 μm),with the mobile phase comprising of acetonitrile-water(containing 0. 1% methanoic acid) flowing at 1. 0 mL/min in a gradient elution manner,and the detection wavelength was set at 270 nm. The results were analyzed by similarity analysis and principal component analysis. RESULTS There were sixteen common peaks in the HPLC fingerprints of thirty batches of aqueous extract of G. elata with the similarities of more than 0. 800,and the counterfeits were different and the similarities were low of 0. 029、0. 016、0. 001、0. 175、0. 041、0. 150、0. 004,which could be used to identify the true and false of G. elata. Eleven different index components were screened by principal component analysis,which showed the fingerprint components of G. elata was consistent. CONCLUSION This stable and reliable method can be used for the quality control of G. elata.
