Extraction and Composition Analysis of Pigment from Jujube Peel

摘要 [目的]以灰枣为原料,初步分析其提取、分离工艺及其活性成分。[方法]通过酸化乙醇提取法对枣皮红色素进行提取,采用单因素试验考察不同pH对枣皮红色素提取效果的影响。在此基础上,利用pH梯度萃取法将枣皮红色素粗提物依次经过5%Na HCO3溶液、5%Na2CO3溶液、1%Na OH溶液进行萃取,得到不同萃取组分。并采用颜色反应和高效液相色谱法(high performance liquid chromatography,HPLC)对萃取物进行初步分析。[结果]通过单因素试验判定出pH=0.5的盐酸乙醇溶液对枣皮红色素提取效果最佳;碱液颜色反应和盐酸共沸颜色反应结果显示,枣皮红色素可能为蒽醌类化合物和多酚类化合物;高效液相色谱法(HPLC)对不同萃取物组分进行初步分析,得出枣皮红色素存在于Na HCO3萃取组和Na2CO3萃取组,并且主要集中于Na HCO3萃取组分。[结论]该研究为枣皮红色素的开发提供理论依据。 [Objective]The grey jujube was used as raw material to analyze its extraction and separation process and its active ingredients.[Method]Acidification of ethanol method was used to extract red pigment from jujube peel.We found the effects of different pH to extract red pigment from jujube peel by single factor experiment.On the basis,through isolation of jujube peel by pH gradient extraction,we could obtain different extraction fractions by the red pigment of jujube peel as followed 5%NaHCO 3 solution,5%Na 2CO 3 solution,1%NaOH solution.High performance liquid chromatography(HPLC)analysis was carried out for the extraction fractions.[Result]pH=0.5 of hydrochloric acid solution extraction effect was best by single factor experiment;Lye color reaction and azeotropic hydrochloric acid color reaction used to analyze the extract fractions,this results showed that jujube red pigment may be anthraquinone compounds and polyphenols compounds;HPLC analysis was carried out for the extraction fractions,the results showed that extract red pigment from jujube peel stored in NaHCO 3 extraction fraction and Na 2CO 3 extraction fraction,and mainly concentrated in NaHCO 3 extraction fraction.[Conclusion]This study provides scientific basis for the red pigment of jujube peel exploitation.
