促红细胞生成素对高糖培养小鼠肾足细胞整合素及整合素连接激酶的影响

摘要 目的:探讨促红细胞生成素(erythropoietin,EPO)对高糖培养小鼠足细胞整合素及整合素连接激酶的影响。方法:体外培养和分化小鼠足细胞,分为对照组(葡萄糖5. 6 mmol/L)、高渗对照(葡萄糖5. 6 mmol/L+甘露醇24. 4 mmol/L)、高糖1组(葡萄15 mmol/L)、高糖2组(葡萄糖30 mmol/L),分别以以0 U/ml、10 U/ml、20 U/ml、50 U/ml浓度的重组人促红细胞生成素(环尔博)进行干预24 h,以Western blot法检测integrinβ_1、ILK、α-SMA表达的情况。结果:western blot结果:在高糖环境中ILK表达上调,当EPO浓度为10 U/ml、20 U/ml、50 U/ml浓度时ILK表达依次下调且有显著意义(P <0. 05)。integrinβ_1表达在高糖环境中表达下调,在高糖环境中当EPO浓度为10 U/ml、20 U/ml、50 U/ml浓度时integrinβ_1表达均上调(P <0. 05),以EPO浓度为10 U/ml时上调明显。α-SMA在高糖环境中表达上调,应用EPO浓度为10 U/ml、20 U/ml、50 U/ml浓度时表达下调,以高糖1组下调明显(P <0. 05)且有显著意义。结论:EPO对高糖环境中的ILK、α-SMA表达上调有抑制作用,对integrinβ_1表达有上调作用。
