页岩气油基钻屑降解菌群的构建及其降解特性 被引量:6

Study on community construction of oil degradation bacteria and degradation characteristics of shale gas oil-based drilling cuttings

摘要 以柴油作为唯一碳源,经过富集驯化,筛选出7株石油烃降解菌单菌,并分别测定其对柴油和油基钻屑的降解效能,利用其中4种菌(K-2、K-3、K-6和K-7)构建复合菌群对油基钻屑进行进一步降解试验。结果表明:(1)K-7对柴油和油基钻屑降解能力最强,降解率分别可达67.5%和28.8%;(2)最佳的复合菌群组合为K-3、K-6和K-7,其对油基钻屑的降解率达到了37.5%,较单菌最优降解率提高约30.2%;(3)气相色谱质谱分析表明,复合菌群能有效降解油基钻屑中的烷烃和芳香烃,对长链烷烃、支链烷烃、烷基萘芳香烃均具有较强的降解能力;(4)生理生化鉴定,初步判断K-6为红酵母菌属(Rhodotorula sp.),K-2、K-3、K-7均为芽孢杆菌属(Bacillus sp.)。 Diesel oil was used as the sole carbon source,and seven strains of petroleum hydrocarbon degrading bacteria were screened out by enrichment and domestication process.The degradation efficiency of diesel oil and oil-based drilling cuttings was determined,and the K-2,K-3,K-6 and K-7 were selected as dosing bacteria to construct community for further degrade the oil-based drilling cuttings by changing the dosing ratio.The results showed that:(1)K-7 had the best degradation ability of diesel oil and oil-based drilling cuttings,which were 67.5%and 28.8%,respectively.(2)The best bacteria combinations was K-3,K-6 and K-7,with the degradation rate of oil-based drilling cuttings was 37.5%,which was about 30.2%higher than that of optimum bacteria.(3)GC-MS results showed that the community could effectively degrade alkanes and aromatic hydrocarbons in oil-based drilling cuttings,and had strong ability to degrade long chain alkanes,branched alkanes and alkyl naphthalene aromatic hydrocarbons.(4)K-6 was identified as Rhodotorula sp.,while K-2,K-3 and K-7 were Bacillus sp.by series of morphological observation as well as physiological and biochemical tests.
