大气细颗粒物暴露对肽基-脯氨酰顺/反异构酶及调节丝氨酸/苏氨酸蛋白激酶B激活磷酸化的影响

摘要 为探讨大气细颗粒物(PM_(2.5))对肽基-脯氨酰顺/反异松酶(PIN1)和丝氨酸/苏氨酸蛋白激酶B(Akt)在肿瘤发生中的影响,揭示PM_(2.5)对人体健康的不利影响,为日后研究提供有益资料。本研究将Wistar大鼠暴露于PM_(2.5)中2~4周,使用MAS-SON染色的方法观察其肺组织胶原产生情况,用Western blot、Real-time q PCR检测肺组织以及H292细胞中PIN1的mRNA和蛋白质变化情况;不同浓度PM_(2.5)(0、100、200μg/m L)处理H292细胞24 h后,使用Western blot方法检测p-Akt的表达情况。结果显示随PM_(2.5)暴露时间的延长大鼠肺组织中胶原产生受到抑制,PIN1的蛋白表达在组织和细胞中随暴露时间的延长和浓度的增加均下调(P<0.05),PIN1的mRNA表达均上调(P<0.05);p-Akt表达随PM_(2.5)浓度的浓度的增加受到抑制(P<0.05)。研究发现PM_(2.5)能够在蛋白水平抑制PIN1以及p-AKT的表达。 In order to investigate the effects of fine particulate matter(PM 2.5)on the development of PIN1 and Akt in tumorigenesis,it reveals the adverse effects of PM 2.5 on human health and provides useful information for future research.Wistar rats were exposed to PM 2.5 for 2~4 weeks.The collagen production in lung tissue was observed by MASSON staining.The mRNA and protein of PIN1 in lung tissue and H292 cells were detected by Western blot and Real-time qPCR.Changes:H292 cells were treated with different concentrations of PM 2.5(0、100、200μg/mL)for 24 hours,and the expression of p-Akt was detected by Western blot.The results showed that the collagen production in the lung tissue of rats was inhibited with the prolongation of PM 2.5 exposure time.The protein expression of PIN1 was down-regulated in tissues and cells with the prolongation of exposure time and concentration(P<0.05),mRNA expression of PIN1.Both were up-regulated(P<0.05);p-Akt expression was inhibited with increasing PM 2.5 concentration(P<0.05).It is found that PM 2.5 inhibits the expression of PIN1 and p-AKT at the protein level.
