摘要 目的:探讨肝细胞生长因子(HGF)对人口腔鳞癌裸鼠移植瘤的作用及其机制。方法:对36只裸鼠皮下接种Tca8113细胞悬液,将32只成瘤鼠随机分为阴性对照组(n=8)、PBS组、HGF 20 ng组、HGF 40 ng组和HGF 100 ng组(n=6),瘤内分别给予不做处理、注射同体积的PBS液、注射HGF 20 ng、HGF 40 ng、HGF 100 ng。经过连续5 d的注射后,测量各组裸鼠肿瘤体积、重量及破溃情况,采用免疫组化染色观察不同剂量HGF对荷瘤裸鼠肿瘤HGF、c-Met及VEGF-C的表达的影响。结果:成功建立口腔鳞癌裸鼠移植瘤模型。分组后各组荷瘤裸鼠体重差异无统计学意义(P> 0. 05),HGF处理后肿瘤体积、瘤重及肿瘤破溃情况差异无统计学意义(P> 0. 05)。免疫组化显示,与阴性对照组和PBS组相比,荷瘤裸鼠肿瘤HGF、c-Met的表达在HGF 40 ng组和HGF 100 ng组明显增高(P <0. 05);荷瘤裸鼠肿瘤VEGF-C的表达在HGF40 ng组最多,与阴性对照组及PBS组相比(P <0. 05)。结论:HGF参与口腔鳞癌肿瘤的生长及破溃,可能通过调控c-Met及VEGF-C的表达起作用。 Objective:To investigate the influence of hepatocyte growth factor(HGF)on oral squamous cell carcinoma(OSCC)in nude mice and to explore its mechanism.Methods:36 nude mice were inoculated with Tca8113 cell suspension for the establishment of xenograft model of human oral squamous cell carcinoma(OSCC)in nude mice.32 nude mice with OSCC were randomly divided into negative control group(n=8),PBS group,HGF 20 ng group,HGF 40 ng group and HGF 100 ng group(n=6).The tumors were given without treatment,injected with the same volume of PBS solution,injected with HGF 20 ng,HGF 40 ng and HGF 100 ng respectively.5 days after continuous treatment,the tumor volume,weight and bursting condition of the nude mice were measured,and the expression of HGF,c-Met and VEGF-C in the tumor was examined by immunohistochemical(IHC)staining.Results:OSCC xenograft model was successfully established in 32 nude mice.There was no significant difference in tumor weight after grouping(P>0.05).There was no significant difference in tumor volume,tumor weight and tumor rupture among groups after treatment(P>0.05).IHC examination showed that compared with the negative control group and PBS group,the protein expression of HGF and c-Met in HGF 40 ng and HGF 100 ng groups increased(P<0.05).The expression of VEGF-C in the HGF 40 ng group was increased(P<0.05).Conclusion:HGF contributes to the growth and ulceration of oral squamous cell carcinoma and it may be mediated by the regulation of c-Met and VEGF-C expression.
