BsNPV和BmNPV重组毒株生物学特性 被引量:1

Determination on biological characteristics of a BsNPV-BmNPV recombinant virus strain

摘要 为了扩大油桐尺蛾核型多角体病毒(Buzura suppressaria nucleopolyhedrovirus,BsNPV)宿主域和实现规模化生产,将其与家蚕核型多角体病毒(Bombyx mori nucleopolyhedrovirus,BmNPV)共同侵染油桐尺蛾幼虫,虫体死亡后收集并纯化获得的病毒(P1)再侵染家蚕,待家蚕死亡后收集并纯化获得的病毒(P2)再侵染油桐尺蛾幼虫,虫体死亡后收集并纯化获得病毒(P3),以此获得能感染油桐尺蛾和家蚕的自然重组病毒新株系,并对该株病毒进行超微结构鉴定和毒力测定(LT_(50))。结果表明,第1代(P1)和第3代(P3)重组病毒的超微形态和大小接近于亲本BsNPV,第2代(P2)重组病毒则与亲本BmNPV更接近,其大小介于亲本BmNPV与P1、P3之间。P3侵染油桐尺蛾幼虫的LT_(50)比亲本BsNPV提前1.79 d。因此,本试验获得的重组病毒对油桐尺蛾的杀虫活性增强。 To expand the host range and facilitate scale production,a Buzura suppressaria nucleopolyhedrovirus(BsNPV)was co-inoculated on B.suppressaria larvae with a Bombyx mori nucleopolyhedrovirus(BmNPV).After the B.suppressaria larvae died,the virus(P1)was collected and infected with B.mori larvae.After their died,the virus(P2)was collected and infested with B.suppressaria larvae.The virus(P3)was collected after the B.suppressaria larvae died.The ultrastructure of recombinant NPV strain was subsequently investigated by scanning electron microscopy(SEM)and transverse electron microscopy(TEM),meanwhile the toxicity to B.suppressaria larvae was measured by LT 50.The results showed that the ultrastructure and size of the first(P1)and third generation(P3)recombinant viruses were close to those of parent BsNPV.The second generation(P2)showed a polyhedron shape,similar with the parent strain BmNPV.However,the P2 virus particles were smaller than BmNPV,but larger than the first or third generation viruses.The LT 50 of P3 recombinant viruses was 1.79 days earlier than that of parent BsNPV in transfection on B.suppressaria larvae.This study has developed a recombinant virus with a promoted insecticidal activity than the parent BsNPV strain.
