摘要
[目的]筛选出利于实验室来源的雨生红球藻营养生长的培养基,优化雨生红球藻内虾青素的积累条件。[方法]选用7种培养基对4株雨生红球藻分别进行培养,筛选出最适于4种藻株营养生长的培养基。探究雨生红球藻在光胁迫、高盐、缺氮3种条件下诱导虾青素积累情况。[结果]4个不同雨生红球藻藻株在配方A培养基中的生物量比其他6种试验培养基的生物量高1.65~7.30倍;在优势培养基配方A中,HP3的生物量最大,可达7.13×106个/m L; HP3在缺氮的胁迫条件下,虾青素产量最高,可达21.05 mg/L,比光诱导获得的最佳虾青素产量高1.07倍,比高盐获得的最佳虾青素产量高1.64倍。[结论]该研究中最适于试验藻株营养生长的培养基是配方A,当胁迫条件为缺氮10%时虾青素积累量最大。
[Objective]Medium for culturing Haematococcus pluvialis originated from laboratory was screened,accumulation conditions of astaxanthin in Haematococcus pluvialis were optimizated.[Method]Seven culture mediums were used to cultivate four kinds of Haematococcus pluvialis,4 kinds of mediums were obtained.Astaxanthin accumulation in Haematococcus pluvialis were exposed under light-induced,high-salt,and nitrogen-deficient respectively.[Result]Biomass about four strains algae in the formula A medium was 1.65-7.30 times of that in the other six mediums,HP 3 gained the largest biomass in formula A,the maximum biomass was up to 7.13×10 6 cell/mL.Under nitrogen deficiency condition,the astaxanthin production was up to 21.05 mg/L,this number was 1.07 times of the best astaxanthin yield obtained by light,and 1.64 times of the maximum astaxanthin yield obtained by high salt.[Conclusion]The most suitable Haematococcus pluvialis medium was formula A and the regnant astaxanthin accumulation condition was 10%in this study.
