光质对苋菜愈伤组织生长及甜菜色素和类胡萝卜素合成的影响 被引量:8

摘要 为探究光质对苋菜愈伤组织中甜菜色素及类胡萝卜素合成的影响,研究蓝光、白光、绿光、红光、远红光、黑暗对苋菜愈伤组织生长状态及类胡萝卜素和甜菜色素含量的影响,并将苋菜愈伤组织中类胡萝卜素提取条件进行优化。结果表明,只有蓝光和白光可以诱导分化出红色愈伤组织,其他光质下苋菜愈伤组织呈黄色或白色;与黑暗相比,蓝光最有利于苋菜愈伤组织中甜菜色素、类胡萝卜素的合成,白光次之,其他光质无法合成甜菜色素;筛选出类胡萝卜素最佳提取条件是:采用丙酮与石油醚比为1∶1混合提取液,苋菜愈伤组织与提取液比为1∶100,提取时间8 h。 For the purpose of exploring effects of light quality on betalains and carotenoids synthesis in amaranth callus,amaranth calli were placed in different light quality including white,green,red,blue,far red and dark,respectively.The extraction condition of carotenoid in the callus was optimized.The results showed that only blue and white light could induce the red callus,and the callus was yellow or white under other light quality.Compared with dark treatment,blue was the most favorable light for the synthesis of betalain and carotenoid in amaranth callus,followed by the white light.The optimal extraction condition of carotenoids was as follows:the ratio of acetone∶petroleum was 1∶1,the ratio of raw material to extraction solution was 1∶100 and the extraction time was 8 h.
